Objective With the development of economy and the improvement ofliving standards, the incidence of diabetes is increasing, significantlydecreased the quality of life of patients with diabetes and shorten its life,diabetic microvascular lesions is an important cause of death in diabetes,the damage to the kidney with diabetes is one of the microvascular diseasecomplications. Oxidative stress in diabetic kidney damage plays animportant role in the process, we study the gastric bypass for early renaloxidative stress of rats with type2diabetes, and preliminary discussion ofits protective effect on renal mechanism.Methods (1) Forty SD rats were randomly separated into normalcontrol group(NC,n=8)and diabetes mellitus model group(DC,n=32),thediabetes model group were injected STZ(35mg/kg),30rates wereconsidered as diabetes mellitus modle. Then they were randomly dividedinto a diabetes control group(DC, n=8),a diabetes sham operation group(DSO, n=8), and a diabetes operation group (DO, n=14);(2)The levels ofglucagon like peptide-1(GLP-1),fasting blood glucose (FBG)wasmeasured before the operation and at1th,2th,4th,8th weeks after the operation;(3) After8weeks of the operation, serum AGE,serumBUN,Cr,24h urine8-OHdG were detected; the kidney weighty index wasmeasured;The activities of antioxidant enzymes in renal including totalsuperoxidedismutase(TSOD), Mn-superoxidedismutase(MnSOD), Cu–Znsuperoxidedismutase(CuZnSOD), Catalase(CAT), Glutathioneperoxidase(GSH-PX) and maleicdialdehyde(MDA) were measured by chromatometry;Changes of MnSOD, CAT, CuZnSOD, GSH-PX mRNA express weredetermined by real-time PCR;Western blot analysis was employed to assessthe protein expression of recepter of advance glycation end products.Results (1) The FBG of diabetes operation group decreased with thetime pass from before the operationt(12.48±1.37)mmol/L to8th weeks afterthe operation (5.45±0.71) mmol/L,;However the FBG of DSO andDCgroup were increase a little,FBG has no obvious change in all groups.(2) The fasting GLP-1of diabetes operation(DO) group increased fr.ombefore the operation (8.12±1.54) pmol/L to8th weeks after the operation(22.35±0.79)pmol/L,(P<0.05), the other groups have no significantlychange.(3) After8weeks of operation,24h urine8-OHdG and MAU,serum AGE, serum BUN,Cr,kidney weighty index (KWI)and MDAincreased significantly in the DC and DSO groups compared with the DOand NC groups(p<0.05). The activities of TSOD, GSH-Px, CAT,CuZnSOD, decreased, significantly in in the DC and DSO groupscompared with NC and DO groups (p<0.05),the MnSOD have no signification different among all groups; the CuZnSOD, GSH-PX mRNAexpress were increased in DC and DSO groups compared the DO andNCgroups(p<0.05) but CAT mRNA express desend(p<0.05),MnSOD haveno change among each group; PAS dyeing indicates that glomerulusextracellular matrix accumulating in the DC and DSO groups,the DO andNC groups have no signification alter. the serum AGE,kidney tissueRAGE protein expression in DC and DSO groups was significantly higherthan that in the NCand DO groups.Conclusion (1) High in fat and high sugar feeding add Strepozotocininjection induction type2diabetic rats model is stable,which is considereda perfect modle of type2diabetes, as the surgery for the treatment ofdiabetes.(2) Gastric bypass operation can protect the early phase of diabetesnephropathy rats.(3) The gastric bypass operation protects the kidneymaybe through increase the GLP-1and re-balancing the oxitive and anti-oxitive system.
