Plasmids transfection and cell culture;Immunoblotting; Embryos in situ hybridization and sections.Results: Clones and their express pattern; Alignment of genes abouttheir amina acid sequences; Temporal and Spatial expression of gens aboutAD during Xenopus laevis early development.Conclusion: We describe the temporal and spatial expression pattern ofAPP, BACE2genes during Xenopus laevis early development and comparethe expression patterns in the developing eye and brain. Alignment of aminoacid sequences shows a high conservation. RT-PCR analysis indicates thatThe Xbace2and Xrcan1transcripts are clearly detected from blastula-stagesto late gastrula stages and its expression remained at a relatively high leveltill neural stages. Strong expression of these genes can be detected at stage30, stage40and stage45. Results demonstrated that during development, thosegenes expressed in a pattern that recapitulates endogenous proteinexpression. Consistent with previous in situ and immunohistochemistry data,we observed those genes expression in multiple tissues and cell types,including the central and peripheral nervous system, neural retina,pharyngeal mesoderm and endoderm and their derivatives (craniofacialstructures, thymus and trachea), cardiovascular system (cardiac outflow tract,carotid arteries, umbilical vessels, sinoatrial node and atrial septum),gastrointestinal system (oral epithelium, stomach, pancreas, mesentery).
