Objiective: To study the influences of IQGAP1on the proliferation,trans-differentiation and migration in alveolar epithelial cells type II.Methods: To interference the expression of IQGAP1in alveolarepithelial cells type II (AEC II) by transfected IQGAP1-siRNA, set upblank control group.1. The effect of IQGAP1on the number of alveolarepithelial cells type II detected by Direct cell count method;2.theproliferation rate of AEC II was analyzed through the methyl thiazolyltetrazolium (MTT) method;3.the role of IQGAP1in the migration of AECII was analyzed by Transwell migration experiment;4.the expression ofAEC II-specific Surfactant-associated protein C(SP-C)and AEC I-specificaquaporin-5(AQP-5) was detected by the Western blotting,to analyzed theeffect of IQGAP1on the differentiation of AEC II.Results:1.Compared with the number of cells in control group,thegroup which interfered by IQGAP1-siRNA was reduced obviously(P<0.01);2.Compared with the cell proliferation rate of control group,the group which interfered by IQGAP1-siRNA was decreased significantly(P<0.01);3.Compared with the control group, siRNA interfered the expression ofendogenous IQGAPl in AEC II, and the cells migration extent decreased byapproximately50%(P<0.05);4.After24hours interfered, the AEC IImarker(SP-C) see a large number expression, and the AEC Imarker(AQP-5) just see a few expression; after48hours showed that theup-regulation of the expression of SP-C and the down-regulation of theexpression of AQP-5,indicating a failure to trans-differentiate to AEC I.Conclusion: IQGAP1plays an important regulatory role in theproliferation, trans-differentiation and migration in alveolar epithelial cellstype II.
