Objective: To research the effects and mechanism ofdihydroartemisinin on modulating UCHL1gene expression in humanprostate cancer cell line PC-3and its mechanism.Methods: Part1, Cell migration and invasion assay were introduced totest the migration and invasion ability of PC-3and DU145cells48hoursafter different concentrations`DHA (0μmol L-1,25μmol L-1,50μmol L-1and100μmol L-1) influences. PCR and WB experiments were used totest to the transcription and expression level of Vimentin and CDH1afterdifferent concentrations of DHA influences.Part2, Prostate cancer PC-3and DU145cells were transplanted into18nude mice to establish the solidtumor mode.These nude mice were randomly divided into3groups with3mice in each group:control group,solvent group (dimethyl sulfoxide1mL/kg weight),DHA group (DHA200μmol/kg weight).The positiveproducts of Vimentin,CDH of PC-3and DU145cells were tested byimmunohistochemical on day13after drug administration.Results: DHA can significantly inhibit cell migration and invasion ofPC-3and DU145cells in different concentrations (25μmol L-1,50μmol L-1and100μmol L-1). And25μmol L-1concentration play the mostobvious function. Compared with the control group, different concentrationsof DHA significantly inhibit Vimentin and promote CDH1transcription andexpression levels.Conclusion: DHA significantly inhibit migration and invasion abilitiesof PC-3and DU145cells in vitro, the underlying mechanism may interferewith the expression level of Vimentin and CDH1proteins associated withDHA.
