There is the larger advantage of the rich eucommia ulmoides resources in China. However, most of the eucommia ulmoides resources have been wasted, because of unreasonable utilization or the lower efficiency of comprehensive utilization. This study based on the plants resources characteristics of eucthe ommia ulmoides in our country, Pinores inoldiglucoside and Eucommia polysaccharide as the purpose components, the concept that high efficient comprehensive utilization of eucommia ulmoides resources was put forward. Its specific content was as follows:the ionic liquids-ultrasonic assisted extraction technology was applied to extracting Pinores inoldiglucoside from eucommia ulmoides skins, and optimized the extraction conditions by response surface method; The Pinores inoldiglucoside was adsorbed and enrichment with macroporous resin from ionic liquid extract of eucommia ulmoides; The ultrasonic-assisted extraction technology was applied to extracting polysaccharide from eucommia ulmoides, and the physical and chemical properties of polysaccharide were studied. The extraction methods and separation technologies of the Pinores inoldiglucoside and Eucommia polysaccharide were studied systematically, and the main content was as follows:(1) A rapid HPLC method which determined the content of Pinores inoldiglucoside was set up. The chromatographic separation was carried out by a Kromasil-C18(5μm,4.6mm×250mm) reversed-phase column, the mobile phase is acetonirtrile-water-phosphoric acid (20:80:0.3, v/v/v). The determination wavelength was228nm. The flow rate was1ml/min, the injection volume was5μL and the column temperature was maintained at25℃, retention time was45min.(2) The ionic liquids with different cations and anions were investigated in this work and the technical parameters including soaking time, liquid-solid ratio, ultrasonic power and time were optimized.1.0mol/L1-butyl-3-methylimidazolium bromide solution as solvent, the soaking time was3h. Base on the single-factor experiments, a three-factor-three-level experiment design had been developed by Central Composite experiment design method. The optimized conditions were as follows:ultrasonic power200W, extraction time was40min, the ratio of liquid to solid was14:1. The actual yield of Pinores inoldiglucoside was3.12mg/g.(3) Seven kinds of macroporous resins were selected for the enrichment and purification of Pinores inoldiglucoside from crude extract, including HPD100A, HPD700, HPD400, HPD750, HPD850, HPD600and HPD826. The optimum purification parameters were established: HPD850resin was screened out from seven kinds of macroporous resin to purify Pinores inoldiglucoside. The best purification process conditions:adsorption temperature was25℃, saturated adsorption time was4hours, the concentration of crude extract was9.66mg/mL, Pinores inoldiglucoside adsorption capacity was15.65mg/g; degreasing agent was20%ethanol solution, desorption time was90min, desorption temperature was25℃, Pinores inoldiglucoside content of desorption extract was14.7mg/g, and the purity was45.8%.(4) Ultrasound-assisted solvent extraction method was applied for extracting the Eucommia polysaccharide. In addition, the technical parameters including soaking time, liquid-solid ratio, ultrasonic power, time and temperature were optimized. Soaking time was2h, ultrasonic power was250W, extraction time was30min, extraction temperature was40℃, and solid-liquid ratio was12:1. The extraction yield of Eucommia polysaccharide was1.9%.(5) The physical and chemical properties of Eucommia polysaccharide were investigated using the multiple indexes. The antioxidant activity analysis and evaluation results of Eucommia polysaccharide showed that under the same concentration, DPPH free radical scavenging ability of Eucommia polysaccharide was higher than that of vitamin C obviously; however, the iron reduction ability of Eucommia polysaccharide was lower than that of vitamin C, and the iron reduction ability of Eucommia polysaccharide was positively correlated with its concentration.
