| Objective:To explore the affect of MacroH2A on cell cycle and cell apoptosis of melanoma cell line B16in vitro, and the regulate function of MacroH2A on cyclinD1ã€cyclinD3ã€CDK4ã€CDK6ã€CDK8; Discuss the molecular mechanisms of MacroH2A suppresses melanoma cell progression. And to know more about the mechanism of the tumorigenesis and developments of malignant melanoma. Discuss a new possible method on targeted therapy for melanoma.Methods:The interference and over-expression vectors of MacroH2A were constructed and then transferred into B16melanoma cells following transfection efficiency analyzed by real-time PCR, western blot and immunofluorescence assays. The apoptosis and cell cycle stage among all the treatment groups. Then cyclin D1, cyclin D3, CDK4, CDK6and CDK8expressions were detected to elucidate the effects of MacroH2A on cell cycle related genes.Results:The results show that over-expression MacroH2A suppresses melanoma cell progression and arrest the cell in G2/M stage. Further, MacroH2A inhibit Cyclin D1, Cyclin D2, CDK6and CDK8expression in B16melanoma cells.Conclution:Taken together, our results demonstrate that MacroH2A as a critical component of chromatin suppresses the development of melanoma cells which results from disordered cell cycle through regulating of cyclin D1, cyclin D3and CDK6genes. |
