Objective:To explore the relationship between Chinese han nationality patients with type1diabetes (T1DM) and IL12B3’UTR polymorphisms.Design:Cross-sectional and case control study.Methods:Polymerase chain reaction combined with restriction fragment length polymorphism (PCR-RFLP) method was used to detect the allele and genotype of IL12B3’UTR polymorphism in615T1DM patients and543cases of normal control group. Patients with type1diabetes can be divided into juvenile onset (JO) and adult onset group (AO) according to whether the patients with onset age is20years old. It can be divided into TA diabetes group and TB diabetes group according to whether the patients are positive for islet autoantibodies. Meanwhile, TA patients can be divided into simple and non-simple GADA antibody positive group on the basis of whether patients are only GADA antibody positive or combined with any other antibodies(IA-2A, IAA, ZnT8A) positive.Results:①Compared with normal control group, there was no statistically significant difference in IL12B3’UTR genotype distribution (P=0.436) and A allele frequency (54.6%vs57.2%, P=0.217) in patients with T1DM.There was no significant difference of genotype distribution in both TA and TB group when compared with normal control.②Compared with normal control,there was no significant difference of genotype distribution (P=0.939) and A allele frequency(54.1%vs54.6%, P=0.939) in T1DM patients (AO group). However, there are significant difference of the AA+AC genotype frequency (85.9%vs79.8%, P=0.029) and A allele frequency(54.6%vs59.8%, P=0.041) in T1DM patients (JO group), when compared with normal control.③Compared with normal control group, there was no significant difference of AA+AC frequency (79.8%vs80.1%, P=0.148) in simple GADA positive patients. However, there are significant difference of AA+AC frequency (79.8%vs84.7%, P=0.006) and A allele frequency (54.6%vs61.2%, P=0.020) in non-simple GADA positive patients. Compared with non-simple GADA positive patients, there are significant difference of AA, AC and CC genotype distribution (P=0.013) and A allele frequency(P=0.009) in simple GADA positive group.Conclusions:After stratification by age and autoantibodies,IL12B3’UTR polymorphism is associated with T1DM in Chinese Han patients, A allele may have genetic predisposition for T1DM.
