Objective:In order to make sure whether Wharton’s jelly could be used as the precursor of cartilage tissue engineering complex, this experiment tries to analyze the organization and the basic composition of the umbilical cord Wharton’s jelly.Method:We obtained the fetus umbilical cord from the operating room of our hospital. Firstly, Structure research:We make the umbilical cord transverse and longitudinal, then served for the HE stain respectively and observe the micro shape and structure of Wharton’s jelly. In addition, obtain some other umbilical cord, and cut it into pieces with5×1×1mm3, then fix it to observe the shape and structure of umbilical cord Wharton’s jelly through the scanning electron microscope (SEM) observation. Secondly, constituent research:the scaffold was prepared and served for the PAS stain, Masson stain and immunohistochemical staining observation. At the same time, we separated the Wharton’s jelly out from outer membrane and arteriovenous of umbilical cord, using the physical and chemical method to separate cells from Wharton’s jelly. We use calculating instrument to count the number of the cells separated from the Wharton’s jelly, then we stain the Mesenchymal stem cells (MSCs) with three different kinds of cell surface antigen labeling:CD29, CD90, and CD45. After that, we calculate the percentage of cells stained with CD29, CD90but without CD45by the help of flow cytometry detector.Results:Firstly, Wharton’s jelly structure:by the help of HE stain and microscope (SEM) observation, we find that the collagen fiber close to the blood vessels mainly the same as the line direction of the vessels, at the peripheral of the umbilical cord, the mainly direction of the collagen fiber surrounds the vessels. The density of the collagen fiber from outside to inside is gradually becoming sparse to dense. Secondly, Wharton’s jelly composition:Wharton’s jelly contains Collagen fiber, GAGs, water and cells. The collagen fiber mainly contains three kinds of form including collagen fiber Ⅰ、Ⅱ and Ⅲ. The density of Wharton’s jelly is2.04×10^6cell/g and the percentage of MSCs is54.67%in all the cells.Conclusion:The organization and the basic composition of the Wharton’s jelly are similar to those of cartilage and can preliminary satisfy the requirements of cartilage tissue engineering. Wharton’s jelly could be used as the precursor of cartilage tissue engineering complex.
