Chemotherapy effectively prevents the growth of tumor cells. But most of chemotherapy drugs are non-targeted and have a variety of adverse effects. Improving the life quality of patients undergoing chemotherapy is an urgent problem to be solved.Sijunzitang, a traditional chinese herb decoction, consists of ginseng, atractylodes, poria and licorice decoction in a certain proportion. It has been used for treating circulation, blood system, metabolic diseases and digestive system diseases for many years in China. The modern pharmacological researchs show that the decoction has the effect on toxicity attenuation.In this study, the toxicity attenuation and chemical composition of Sijunzitang extracts were studied. The effect of Sijunzitang extracts as adjuvant cancer treatment during chemotherapy with epothilone B was identified on ICR mice. The Sijunzitang extracts were divided into lipsoluble part and water soluble part by extracting with n-butanol. Water soluble part of Sijunzitang extracts were further divided into polysaccharide A part and polysaccharide B part by precipitating using60%ethanol. The content of flavonoids, saponins and sugars were measured by colorimetric method. The UPLC-PDA was used to determine the content of liquiritin, panaxsaponin and glycyrrhizic acid. The structure of polysaccharide B part was primarily analyzed by1H-NMR、13C-NMR and HSQC. Quantitative analysis of sugar residues by1H-NMR was applied to polysaccharide B part of Sijunzitang.ICR mice were given epothilone B, a potential cytotoxic agent used for cancer chemotherapy, to establish the animal model. When mice received a combination of epothilone B and Sijunzitang extracts, their survival rate had been improved compared with that mice treated with epothilone B. Sijunzitang extracts also improve the leukocyte, lymphocyte, thymus index, spleen index and bone marrow DNA and reduce the number of enterobacteria of receiving epothilone B mice.Sijunzitang lipsoluble part mainly compositions, flavonoids and saponins, were determined by colorimetric method. The content of total flavonoids and total saponins was7.89%and24.16%, respectively. The content of liquiritin, ginseng saponin Rg1and glycyrrhizic acid were determination by UPLC-PDA. The results showed that the content of liquiritin, ginsenoside Rg1and glycyrrhizic acid was13.12mg/g,6.46mg/g and13.00mg/g, respectively.The content of polysaccharide in part B extracts is determined by phenol-sulfuric-acid and n-hydroxydiphenyl and showed that the content of neutral sugar and uronic acid were68.90%and2.99%, respectively. The monsaccharides were further analyzed by HPLC after part B was hydrolyzed and showed that it mainly composed of xylose, fructose and glucose.The residues of polysaccharide in part B extracts was primarily analyzed using1H-NMR,13C-NMR and HSQC.It mainly composed of'l)-β-D-Glcp-(6','1)-α-D-Glcp-(6','1)-α-D-Glcp,'2)-β-D-Fru,'1)-α-L-Araf and'1)-α-L-Araf-(3,5'. The'1)-α-D-Glcp-('1,1')-β-D-Glcp-(6',and'1)-α-D-Glcp were implied coming from all of the four herb consist of Sijunzitang.'2)-β-D-Fru was derived from gigseng, licorice and atractylodes.'1)-a-L-Araf-(3,5'and'1)-a-L-Araf were from atractylodes.The content of sugar residues was further analyze by1H-NMR and showed that the main sugar resides,'1)-α-D-Glcp, were derived from gigseng, licorice and atractylodes.
