Objective:To study the effects of NgR-siRNA and NT-3gene trans fection on spinal cord injury reparation in rats.Method:108SD rats were chosen,male or female,of which weights varied form200-250g. These rats were completely randomly divided into shame group (n=27), operation group (n=27), NgR-siRNA group (n=27) and NgR-siRNA+NT-3group (n=27). Each group was divided into9subgroups, each of which included3rats, in the light of the time after operation:12h, Id,2d,3d,5d,7d,14d,21d and28d. The hemisection modelof spinal cord was made by direct cut at the level of T8. Cell suspensionwas injected into the injured spinal cord area from upper and lower ends within5minutes with a micro syringe, which was fixed for5minutes.Besides, medical biological glue was also used to prevent effluence of suspension.Recovery of motor function in rats was assessed according to these time points mentioned above, using samples of spinal cord were collected. The effect of NgR-siRNA and the combination of NgR-siRNA and NT-3gene trans fection on the reparation to spinal cord injury was estimated, using QPCR, Wb to detect the change of expression of bFGF, NGF, MBP. The data were described with mean and standard deviation,do analysis by variance.Comparison between the two was made with SNK.Result:1,QPCR shows that NgR-siRNA+NT-3group has higher level of expression than NgR-siRNA and control groups;NgR-siRNA group has higher level of expression than the operation group.2,Wb shows that NgR-siRNA+NT-3group has morehigher level of bFGF protein expression than shame group duing the time of three weeks,there is no statistical difference at the fourth week;There is no statistical difference on the level of NGF protein expression among experimental groups during the first three days;All experimental groups have more higher level of NGF protein expression than shame group from the first week to the third week;NgR-siRNA+NT-3group has higher level of MBP protein expression than NgR-siRNA and control groups;NgR-siRNA group has higher level of MBP protein expression than the operation group.Conclusion:1,NgR-siRNA neural stem cells which transplanted into rats can promote injured spinal cord reparation;2, NgR-siRNA+NT-3neural stem cells which transplanted into rats can promote injured spinal cord reparation,furthermore the effects of NgR-siRNA+NT-3neural stem cells are superior to NgR-siRNA neural stem cells.
