The Expression And Significance Of BCRP In Peripheral Blood Monocyte/macrophage Of Rheumatoid Arthritis

Objective:To detect the expression of Breast cancer resistance protein(BCRP) in peripheral blood monocyte-macrophage of rheumatoid arthritis(RA). Whether BCRP in peripheral blood confer immune inflammation and multidrug resistance of RA.Method:Patients were divided into RA untreated group13cases, treatment group15cases and refractory group24cases,based on the standard of enrollment.Contral group have13cases.Peripheral blood monocyte-macrophage cell isolation by immune magnetic bead separation,and qPCR detect the expression of BCRP mRNA in peripheral blood monocyte/macrophage. SPSS19.0statistical software was used to analyze the differences between the data of each group, and the relevance of BCRP expression levels with clinical parameters.Result:1.compared to contral group, expression levels of BCRP mRNA in Peripheral blood monocyte-macrophage of total RA groups are significantly invresased(p=0.016, p=0.022, p=0.000; respectively,p<0.05).2.there are no significant differences among untreated group、treatment group and refractory group of RA:compare RA untreated group and treatment group (Z=-0.299, p=0.765, p>0.05); compare RA untreated groupand RA refractory group (Z=-1.443, p=0.149, p>0.05); RA treatment groupand RA refractory group(Z=-1.495, p=0.135, p>0.05).3.BCRP mRNA levels in total RA group showed no correlation with course of disease、the numbers of swollen and tendness、VAS score、ESR、CRP、RFIgG、RFIgA、RPIgM anti-CCP and DAS28score (p>0.05)Conclusion:1.BCRP in Peripheral blood may confer immune inflammation of RA2.BCRP in Peripheral blood may have not association with the treatment of DMARDs.
