Objective:To detect the expression levels of eIF3a and eIF3a gene polymorphisms to study of relationship between eIF3a gene polymorphism and platinum sensitivity in ovarian cancerMethods:q-PCR technology was used to detect the expression levels of eIF3a in52case of ovarian cancer and10case of normal ovarian, to compare eIF3a mRNA expression differences between ovarian cancer tissues and normal ovarian tissue, among varying degrees of differentiation of ovarian cancer and between the drug group and sensitive ovarian cancer group. MassARRAY technology was used to detect the genotyping of eIF3a polymorphic loci in57case of ovarian cancer, to study of relationship between eIF3a gene polymorphism and platinum sensitivity in ovarian cancer. The dates were analyzed by SPSS19.0Results:1. The expression levels of eIF3a in cancer tissues is higher than in normal tissues (0.33±0.16,0.20±0.05respectively, P <0.05).2. The expression levels among high, moderate and low differentiation of ovarian cancer (0.28±0.14,0.32±0.17,0.34±0.17respectively) were similar (P>0.05). 3. The expression levels in platinum drug-sensitive group and drug-resistant group of ovarian cancer (0.32±0.18,0.35±0.16respectively) were similar (P>0.05).4. The distribution frequency of AA genotype in the rs3824830loci of eIF3a was44.0%in platinum-resistant group and56.0%in the platinum-sensitive group, GA genotype was33.33%and66.67%respectively, GG was77.78%and22.22%respectively. Rebalanced based on the age and tumor stage, there was no significant difference (P>0.05) among these three genotype groups.5. The distribution frequency of CC genotype in the rs77382849loci of eIF3a was42.85%in platinum-resistant group and57.15%in the platinum-sensitive group, TC genotype was57.14%and42.86%respectively. Rebalanced based on the age and tumor stage, there was no significant difference (P>0.05) between these two genotype groups.6. The distribution frequency of GG genotype in the rs10787899loci of eIF3a was60%in platinum-resistant group and40%in the platinum-sensitive group, GA genotype was40.74%and59.26%respectively, AA genotype was33.33%and66.67%respectively. Rebalanced based on the age and tumor stage, there was no significant difference (P>0.05) observed among these three genotype groups.7. The distribution frequency of GG genotype in the rs3740556loci of eIF3a was100%in both platinum-resistant group and platinum-sensitive group.Conclusion:1. eIF3a could be ovarian cancer biomarkers for diagnosis.2. For these patients with serous ovarian cancer, mutant genotype was detected in rs3824830,rs77382849and rs10787899loci in eIF3a gene except the rs3740556loci, but no significant correlation was found between these gene types and the chemo sensitivity of platinum.
