Objective:Transient receptor potential canonical-3is a nonselective cation channel and widely expressed in biological membrane of mammal cells.TRPC3is thought to play a role in cellular physiological activities and also be involved in the development of tumor.To elucidate its role in hepatocarcinogenesis, we examined the expression of TRPC3in human HCC samples and adjacent tissues.Methods:Tissue samples and clinical data of this study were obtained from54patients with Primary Hepatocellular Carcinoma(HCC) who underwent hepatectomy between2008and2011in Department of Hepatobiliary and Pancreatic Surgery department of the2nd xiangya Hospital, Central South University. The expression of TRPC3was assessed by immunohistochemistry and Western Blot in this study. Furthermore, the correlations between expression of TRPC3and clinicopathological parameters were analyzed.Result:1.Immunohistochemistry:The majority of well differentiated HCC showed high TRPC3expression(22/30,73.3%),while much fewer poorly differentiated HCC showed high TRPC3expression (6/24,25.0%).2.Western Blot:The higher expression of TRPC3was detected in tumor tissues compared with adjacent tissues.3.Clinicopathological evaluation indicated a significant correlation between TRPC3expression and histopathological differentiation (P<0.05). Conclusions:Our results indicate TRPC3may play a role in hepatocarcinogenesis.
