Objective To investigate the effect of HMGB1on the autophagy of HepG2cells.Methods Hepatocellular carcinoma cell line HepG2cells were transfected transiently with HMGB1-pcDNA3.1、HMGB1siRNA and treated with recombinant HMGB1(40ng/ml); western blotting was used to detect the expression of HMGB1and the ratio of LCII/LCI; electron microscopy was used to detect the autophagosome.Results:1. After transfection with HMGB1-pcDNA3.1, the expression of HMGB1in HepG2cells increased, while the siRNA group decreasd.2.The ratio of LCII/LCI increased in the HMGB1-pcDNA3.1group and r HMGB1group, autophagosomes were obviously visible under the electron microscopy, while in the siRNA group, the expression of HMGB1and the ratio of LCII/LCI decreasd.Conclusion:HMGB1can induce the autophagy of HepG2cells. |
