Objective:Observing the change of ratio of Th9cells and IL-9level in peripheral blood between acute phase and recovery phase of patients with GBS and the IL-9level in cerebrospinal fluid of patients with GBS in acute phase, analyzing the correlation between several indicators, and discussing the effect and clinical significance of Th9cell in the process of GBS.Methods:Peripheral blood and the cerebrospinal fluid of18patients with GBS during the acute phase (within2weeks) and peripheral blood of patients with GBS during the recovery phase(4weeks later) are collected. The peripheral blood of15normal controls(NC) and the CSF of patients with other noninflammatory neurologic diseases (5patients with primary headache and3patients with normal pressure hydrocephalus, named other noninflammatory neurologic diseases group,ONND) are also collected. The ratio of average age and sex between GBS group and other two groups show no statistical significance. The ratio of Th9cells (labeled as CD3+CD8"IL-9+) of peripheral blood of GBS group in the acute phase and recovery phase and the ratio of Th9cells of peripheral blood of normal controls are detected by flow cytometry. The IL-9level of peripheral blood of GBS group in the acute phase and recovery phase and the IL-9level of peripheral blood of normal control group are detected by ELISA. The IL-9level of CSF of GBS group in acute phase and the IL-9level of CSF of ONND group are also detected by ELISA. The IgG level of CSF and Hughes scores of patients with GBS in acute phase are recorded. The correlation between several indicators is analyzed.Results:(1)The ratio of Th9(0.79±0.39%vs0.20±0.07%) and IL-9(4.70±1.01pg/ml vs3.32+0.08pg/ml) level in blood of the GBS group in the acute phase are higher than that of normal control group, the difference is statistically significant (P<0.01). The ratio of Th9(0.79±0.39%vs0.45±0.27%) and IL-9level (4.70±1.01pg/ml vs3.67±0.64pg/ml) in the acute phase of GBS group are higher than that in recovery phase of GBS group, the difference is statistically significant (P<0.01). The ratio of Th9(0.45±0.27%vs0.20±0.07%) in recovery phase of GBS group is higher than that of normal control group, the difference is statistically significant (P<0.01).The IL-9level (3.67±0.64pg/ml vs3.32±0.08pg/ml) in recovery phase of GBS group is higher than that of normal control group, but it is not statistically significant (P>0.05). The ratio of Th9(1.13±0.28%vs0.62±0.32%) and IL-9level (5.57±0.78pg/ml vs4.27±0.83pg/ml) of severe GBS group in acute phase are higher than that of mild GBS group, the difference is statistically significant (P<0.01). The IL-9level (10.07±2.20pg/ml vs4.01±0.30pg/ml) of CSF of the GBS group in acute phase is higher than that of other noninflammatory neurologic diseases group, the difference is statistically significant (P<0.01).(2) The ratio of Th9in blood of patients with GBS in acute phase is positively with the Hughes scores(r=0.706, P<0.01), IgG of the CSF (r=0.690,P<0.01). The IL-9level in blood of patients with GBS in acute phase is positively with the Hughes scores (r=0.643, P<0.01), IgG of the CSF (r=0.496,P<0.05). The IL-9level in blood of patients with GBS in acute phase is positively with the IL-9level of CSF of the patients with GBS (r=0.486,P<0.05).Conclusions:(1)Th9cell may participate in the pathogenesis of the GBS by releasing IL-9.(2) The ratio of Th9and IL-9level in peripheral blood of patients with GBS in acute phase are positively with the severity of disease.
