Objective:To study the expression of vascular endothelail growth factor (VEGF) and oncostatin M(OSM) in airways and lungs of mice with asthma;To observe whether VEGF and OSM participate in the airway remodeling of asthma mice;To observe whether the inhaled budesonide,will affect the VEGF and OSM level in lung tissue of asthma mice.Methods:Mice were sensitized and challenged with ovalbumin (OVA) to establish the asthmatic model. Forty-five BALB/c SPF female mice were randomly divided into a control group, an asthma group, a budesonide intervention group, with15mice in each group.(1)control group:sensitized and challenged with saline.(2)asthma group:first intraperitoneal injection ovalbumin and aluminium hydroxide suspension solution to sensitization, then the ovalbumin solution atomization excitation;(3)BUD group:after sensitization, each excitation prior to atomization inhaled budesonide. Within24hours after the last challenge,performed to detect the pathological changes of each group mice airways and lungs tissue were observed by HE and Masson staining. The bronchial smooth muscle thickness were measured by using computer-assisted image analysis system. The expression of vascular endothelail growth factor (VEGF) protein and oncostatin M(OSM) protein were detected by immunohistochemistry (IHC). The mRNA levels of vascular endothelail growth factor (VEGF) and oncostatin M(OSM) were identified by RT-PCR.Results:1.Pulmonary histopathology changes:HE staining:compared with CTRL group and BUD group, OVA group have higher airway epithelial thickening, smooth muscle cell proliferation, hypertrophy, epithelial cell necrosis, vascular proliferation, peripheral airway and vascular more inflammatory cell infiltration, alveolar septal structure disorder and alveolar wall visible fracture; the above performace in BUD group of mice is significantly lower than OVA group; In group CTRL, without the infiltration of peribronchial inflammatory cells,and the alveolar walls are intact, there have statistical significance between the three groups on the vessels number and the airway epithelial thicken(P<0.05).Masson stained lungs of asthma group demonstrated lots of collagen deposition around the airway and vessel. These changes were significantly reduced in BUD group.The CTRL group have not such changes.2. Immunohistochemistry was used to detect the expression of VEGF protein and OSM protein in three groups of mice:VEGF protein expression in OVA group, compared with BUD group and CTRL group, were increased, and the difference was statistically significant (P<0.05); BUD group compared with CTRL group, the expression of VEGF protein increased, the difference was statistically significant (P<0.05); the OSM protein expression in OVA group compared with the BUD group^CTRL group, were increased significantly, the difference was statistically significant (P<0.05), BUD group compared with CTRL group, the expression of OSM increased, the difference was statistically significant (P<0.05).2.RT-PCR to detect the expression of VEGF and OSMmRNA in three groups of mice:the expression level of VEGFmRNA and OSMmRNA in airway and lung tissue in OVA group is higher than those in CTRL group and BUD group, the difference is statistically significant (P<0.05).3.the result of correlation analysis:the expression of OSM and VEGF protein in airway and lung tissue of mice is positively correlated (r=0.83,P<0.05)。4.Results of correlation:There was correlation between the protein expression of VEGF and OSM in asthma group(y=0.83, p<0.05). Conclusions:1. The expression of VEGF and OSM in the airways and lung tissue of the asthma mice are increased, budesonide intervention can make its expression decreased.2.VEGF and OSM are involved in airway remodeling process of asthma, a linear correlation between them.3. budesonide can affect the airway remodeling of asthma by reducing the expression of VEGF and OSM.
