OBJECTIVE:To study the changes of PV and NeuN positive neurons of human visual cortex with aging, providing morphological basis for neurodegenerative changes.METHOD:(1) We collected17cases of human brain specimens, visual cortex were separated from brains and embedded in protocol and cut into slices. Immunohistochemistry was carried out to observe PV, NeuN immunoreactive neurons under optical microscope. Calculate the cell density and integral optical density value, analyse data.(2)6E10immunohistochemical staining was performed on the above-mentioned17cases of visual cortex adjacent piece, and analysis correlation among6E10,PV and NeuN.RESULTS:The PV positive neurons were mainly distributed in the external pyramidal layer, external granular layer and inner granular layer, their cell bodies were oval, spindle or irregular shape, their dendritic were clear, and their number was gradually decreased with aging. NeuN positive neurons were distributed in all layers of the visual cortex, their nuclei were stained deeply, cytoplasm and dendrites were stained lightly. The number of PV and NeuN immune positive neurons was decreased significantly in the visual cortex which expressed6E10.CONCLUSION:With aging, the number of PV and NeuN immunoreactive neurons in the visual cortex of human was decreased, and it was decreased obviously in specimens which expressed6E10immunoreactive plaques.
