Objective:This study was designed to explore the antineoplastic function and the molecular mechanism of the native Shiraia bambusicola. The Gastric carcinoma cell BGC823,Lung carcinoma cell A549, Hepatoma carcinoma cell HepG2and Cervical carcinoma cell Hela were used as cell models.The study was designed to observe the effects of different Shiraia bambusicola components on cellular morphology and proliferation, to test the change of cellular apoptin,and to analyze the molecular mechanism of those Shiraia bambusicola components to those cell models. This research will support a theoretical basis for utilizing Shiraia bambusicola components effectively.Methods:(1) Growth inhibition of Shiraia bambusicola components to those four model cells were measured by MTT assay.(2) Morphological changes of BGC823was observed by Hoechst fluorescence staining.(3) Apoptosis rate of BGC823cell and cell cycle condition were anlysed by FCM.(4) Western blotting was applied to analyze the expression of PARP and FLIP.Results:(1) The Shiraia bambusicola component1and3can dramatically inhibit the growth of BGC823; the effect which to other three tumor cells is not obvious.(2) The Shiraia bambusicola component1and3can promote the apoptosis of BGC823.(3) The Shiraia bambusicola component3can prevent the cell cycle of BGC823in S period and promote the apoptosis in high concentration (4) The Shiraia bambusicola component3stimulated the expression of PARP in certain concentration.(5) The Shiraia bambusicola component3inhibited the expression of FLIP in certain concentration.Conclusion:(1) The Shiraia bambusicola component1and3have strong pharmacological effects.(2) The Hoechst33324fluorescence staining shows that the Shiraia bambusicola components may inhibit the growth of BGC823by stimulating apoptosis.(3) When component1and 3are involved in cell proliferation, some proteins, like PARP, FLIP, paly important roles.(4) The component2and4have no effect on this experiment.
