| This paper is composed of four chapters. The first chapter did a review of varieties of cultivated Cucurbita moschata Duch.(pumpkin) and the biological activity of it. The second chapter investgated antioxidant activity and a-glucosidase inhibitory activity of six varieties of C. moschata(Jingou, Tianmian, Riben, Chaotianmiben, Miben, Liaoningxinming) cultivated in vitro. The third chapter studied the effect of the extract of various varieties of C. moschata(Jingou and Tianmian)against alloxan-induced diabetic mice. The fourth chapter discussed the study of volatiles components of various varieties of C. moschata(Jingou, Tianmian, Riben, Chaotianmiben, Miben, Liaoningxinming and Xiehuamian).Chapter I Research of varieties of cultivated Cucurbita moschata Duch.(pumpkin)This chapter illustrate the research of C. moschata from the use of it, the biological activity of it, the technology of cultivating and managing it. The use of C. moschata can include the edible, the watch, the use of seed of it.The biological activity of C. moschata including antioxidant activity, hypoglycemic activity, antineoplastic activity and other biological activities.Chapter II Antioxidant and a-Glucosidase inhibitory activity of six varietiesof Cucurbita moschata Duch.cultivated in vitroAntioxidant activity of different extracts of C. moschata was evaluated by scavenging1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and [2,2’-azino-bis (3-ethylbenzothiazoline)-6-sulphonic acid] diamonium salt (ABTS) radical in vitro.The results showed that the DPPH radical scavenging(1%=38.99%), ABTS radical scavenging (IC50=20.90μg/mL), and ferric reducing antioxidant power(TEAC=459.40±10.44μmol/g) of ethyl acetate extract of chaotianmiben C. moschata were higher than that of others. But it was still lower than that of positive control BHT(I%=93.93%,IC50=7.72μg/mL, TEAC=1581.68±97.41μmol/g).By established a-glucosidase inhibitory model in vitro, a-Glucosidase inhibitory activity of C. moschata was screened by96-microplate method in vitro. Results indicated that among which petroleum ether extract of C. moschata(Jingou) had the highest inhibitor activity of a-glucosidase(IC50=143.91μg/mL), which was lower than that of Acarbose(IC50=1103.01μg/mL).Chapter Ⅲ The hypoglycemic effect of Cucurbita moschata Duch.(Jingou and Tianmian) This chapter mainly introduced the hypoglycemic effect of extracts from C. moschata.The experiment showed that Ethyl acetate extract and methanol extract from two varieties of C. moschata(Jingou and tianmian) could significantly decrease fasting plasma glucose(P<0.01and P<0.05, respectively), but the effect to postprandial blood glucose was not obvious. All of the extract could obviously improve liver glycogenin liver(P<0.001, P<0.01and P<0.05, respectively), significantly increase the level of SOD in mice serum and reduce the level of MDA. But the extracts had no function to TC and TG of diabetes model mice.Chapter IV Study of volatiles componentsof various varieties of Cucurbita moschata Duch.The volatile constituents of various varieties of C. moschata were analyzed by head-space solid micro-extraction, coupled with GC-MS and Kovats indices for the first time. The results showed that the volatile constituents were different between the same place of various varieties of C. moschata.The volatiles of two similar species were analyzed by head-space solid micro-extraction, coupled with GC-MS and Kovats indices for the first time. Results found the volatiles of two similar species was very similar. |
