Objective To over-express Cystathionine-β-Synthase(CBS) gene,andinvestigate its effect on APP/Aβ and BACE-1metabolic in cortical neuronsMethed The Cortical and hippocampus neutons were infected with thepackaged lentivirus,And it was designated as CBS group.Cells infected withCBS-NC was designated as NC group and not-infected cell was CON group.The levels of CBS mRNA were detected by real-time quantitative PCR.TheCBS APPand BACE-1protein was detected by western blot and the levelsof Aβ40were measured by ELISA(enzyme-linked immunosorbent assay).Results After the infection of CBS mRNA recombinant lentivirus.theexpression level of CBS mRNA in the cortical neurons increasedsignificantly versus NC and CON groups(P<0.01).The updated expression ofCBS proteins was consistent with CBS mRNA.The levels of Aβ40、APPandBACE-1expression were decreased significantly. ConclusionThe overexpression of CBS Mediated by Lentivirus mRNA effectivelyincreases the levels of Aβ40、APPandBACE-1overexpression in thecortical neurons.
