ObjectiveTo investigate the effects of exogenous fibroblast growth factor(FGF-15) on liverregeneration in rats with biliary obstruction.Materials and Methods48Healthy male SD rats were divided randomly into either a FGF-15treated group,obstructive jaundice(OJ) group, or sham operation(SO) group. Rats in FGF15treatedand OJ group underwent ligation of the common bile duct, while only the SO grouphad division of the common bile duct. Rats in the FGF-15treated group received dailyintravenously of FGF-15at a dose of150ug/kg after the operation. Serum and liversamples were retained.The serum levels of total bilirubin(TBIL), directbilirubin(DBIL), alanine aminotransferase(ALT) and aspartate aminotransferase(AST)were measured.HE staining of liver specimens and light microscopy of liver cells inmitosis case.Quantitative fluorescence real-time PCR was performed to detect hepaticHGFmRNA expression. Immunohistochemistry for hepatic proliferating cell nuclearantigen(PCNA) labeling index was used to evaluate and epitope of FGF-15.Results1. serum levels of ALT and ASTCompared SO group with FGF-15treated group and OJ group after days3and7ofobstructive jaundice, FGF-15treated group and OJ group’s serum levels of TBIL、DBIL、AST、ALT were significantly higher(p<0.05). Compared to the OJ group afterdays3and7of obstructive jaundice, the FGF-15group had significantly lower serumlevels of ALT and AST(p<0.05). But there was almost no increase of serum levels ofTBIL and DBIL(p>0.05). Compared days7with days3of FGf-15group, serumlevels of AST、ALT、DBIL、TBIL were also significantly increased(p<0.05). 2. Evaluation of postoperative liver regeneration in ratsHepatic HGFmRNA, PCNA labeling index and hepatocyte nuclear mitotic in OJgroup rats and FGF-15treated group rats were significantly higher than SO group ratsat days3and7after the operation(p<0.05). Compared to the OJ group, the FGF-15group had significantly high levels of hepatic HGFmRNA, PCNA labeling index andhepatocyte nuclear mitotic at days3and7after the operation(p<0.05). Compareddays7with days3of FGf-15group rats, hepatic HGFmRNA, PCNA labeling indexand hepatocyte nuclear mitotic were also significantly increased(p<0.05).Conlusions1.Exogenous fibroblast growth factor15may enhance obstructive jaundice ratsimpaired liver function;2.Exogenous fibroblast growth factor15may enhance hepatic HGFmRNA expressionand promote liver regeneration in rats with biliary obstruction.
