Objective: To investigate the protective effect of free triiodothyronine on liverinjury induced by D-GalN in rats and impact on tumor necrosis factor.Methods::1.The effects of different doses of D-galactosamine rats: Male SD rats of clean grade wererandomly divided into4groups and treated as follows: the experiment group weregiven200mg/kg D-GalN,400mg/kg D-GalN,800mg/kg D-GalN throughintraperitoneal injection respectively, while the control group was given normal salineby intraperitoneal injection. Blood samples were gathered from celiac artery48hourslater,then all rats were killed and artery blood sampl and liver tissues were collected.Automatic biochemical analyzer levels of ALT, automatic electrochemiluminescence detectedFT3, FT4and detection of TNF-αELISA in blood were detected,as well as, HEstaining were performed in liver tissues.,optical microscope and pathological examination2. The exogenous FT3intervention: Male SD rats were randomly divided into5groups and treated as follows: the1group were injected with T30.01mg/kg and400mg/kg D-galactosamine after48hours,the2group were injected T30.01mg/kgand D-galactosamine400mg/kg at the same time, the3group only dued with400mg/kg D-galactosamine, the4only only dued with T30.01mg/kg,the controlgroup was injected with normal saline. All injections were finished by intraperitonealinjection, five groups of rats were killed at48hours later. Blood samples weregathered from celiac artery48hours later,then all rats were killed and liver tissueswere collected. Automatic biochemical analyzer levels of ALT, automaticelectrochemiluminescence detected FT3, FT4and detection of TNF-αELISA in blood weredetected,as well as, HE staining were performed in liver tissues.,optical microscopeand pathological examination. Result:1, Serum level of ALT in rats with injection of D-galactosamine increasedsignificantly higher than the control group after48hours(P<0.01), the different dosegroups of rats were significantly different (P<0.01), while level of FT3, FT4decreased obviously compared with the control group(P<0.01), different dose group ratshave significantly difference (P<0.01), TNF-αincreased significantly higher than thecontrol group(P<0.01), the differences between different dose group rats weresignificant (P<0.01).2, The supplement of exogenous FT3(either given or is given at the same time),could reduce the serum level of ALT and TNF-αin rats, the difference wasstatistically significant (P<0.01).Conclusion:1, Acute liver injury exist after D-galactosamine injection, injury degree increaseswith the increased dose.2, Rats were euthyroid sick syndrome of acute liver injury model, and with the degree of liverdamage increased, FT3, FT4decreased more significantly, serum TNF-α increased, with thedegree of liver injury aggravated, elevated levels of more obvious3, The supplement of exogenous FT3can reduce the acute liver injury in rats andreduce serum TNF-αlevel.
