Background:In last few years, the incidence and death proportion of cancer in the worldwide continues to rise,Cancer has become the first cause of death in humans. With the rapid development of medical science andtechnology, the level of diagnosis and treatment for cancer improve significantly, but still can’t meet theneeds of humans and the clinical. Numerous studies show, tumor markers has important significance in theearly detection, early diagnosis, clinical stage, assessing the efficacy and prognosis of tumor. Detecting thetumor markers of human blood conveniently、fast、accurately in the early stage is easy to detect the cancerpatients and conduct appropriate intervention for them, can help to improve prognosis. Now, the clinicaldetection of tumor markers are mainly performed in central clinical laboratory requiring professionaloperating、cumbersome instrumentation、long turn-around time and expensive, this is not convenient tomonitor for tumor markers in time.Lateral flow immunoassay strip is a kind of new rapid immunoassay technique developed fromthe end of last century. It has the advantages of rapidness、preciseness、convenient、fast、economy and canbe detecting by the bed or the scene of sampling. LFIAs have acquired broad penetration in a variety ofterritories such as medical diagnosis and therapeutic monitoring. Nevertheless, the optical lateral flowimmunochromatographic assay upon the colloidal gold only can play as qualitative or half-quantitativedetection, and there still need further improvement in terms of sensitivity and precision. Recently, magneticsignal detection based on magnetic nanometer material becomes the research hot of quantitativeimmunochromatographic technique.Objective:To build the strip examination system of magnetic lateral flow immune chromatography.Detection system with superparamagnetic nanoparticles as markers, achieve detecting tumor markers ofcarcinoembryonic antigen conveniently、fast、accurately in the early stage.Methods:1. Firstly, connecting the carboxyl-modified magnetic nanoparticles with antibody using EDC/NHS as activator make into immunomagnetic nanoparticles, then characterize them by scanningelectron microscope、infrared spectroscopy and BCA protein quantitative detection kit.2. By optimizing the system of immunomagnetic beads preparation、the parts of magnetic lateralflow immunoassay strip, establish the strip for detecting tumor markers of carcinoembryonic antigen andtest the properties of strip.3. Detect one thousand and twenty cases of clinical serum samples with this strip detectionsystem and electro-chemiluminescence immunoassay commonly used in clinical, then analyse the testresults comparatively and verify the application value further of this strip detection system.4. Statistical analysis was conducted with the statistical software(SPSS),version17.0. Correlationanalysis、t test and χ2test was performed for analysing the precision of this strip detection system.Results:1. The immunomagnetic beads made by using EDC/NHS as activator has uniform particle sizeand good dispensability and is adapted to establish lateral flow immunoassay system.2. The optimal diameter of superparamagnetic nanoparticle used for establishing lateral flowimmunoassay strip is100nm. During the preparation of immunomagnetic nanobeads,40μg antibody addedto1mg magnetic nanobeads is the optimal and the best type of nitrocellulose membrane is Sartourius CN140. The entire testing process can be finished in15minutes.3. The qualitative detection range of this magnetic lateral flow immunoassay strip detectionsystem for CEA standards is from10ng·ml-1to1000ng·ml-1, the quantitative detection range is from0.1ng·ml-1to1000ng·ml-1and that the stability、specificity and repeatability are all good.4. The correlation of clinical serum samples test results with this strip detection system andelectro-chemiluminescence immunoassay is well(P<0.05).The batch to batch reproducibility of this stripdetection system is good(P>0.05)and that has high precision and stability.Conclusion:In this study, we establish CEA magnetic lateral flow immunoassay strip with superparamagneticnanoparticles as markers, can be used for quantitative and visual detection of serum tumor markerscarcinoembryonic antigen with magnetic assay reader, realize the goal of detecting tumor markers fast、 accurately、conveniently. It can reduce testing costs and improve work efficiency and has broad applicationprospects. Simultaneously, it also could provide theoretical and experimental basis for prophase screeningand therapeutic effect monitoring of cancer.
