Treatment of bone defect is one of difficult subjects in orthopedic surgery. Bone Marrow-derived Mesenchymal Stem Cells (BMSCs) from Mongolian sheep as seed cells, bone allograft as a cell carrier, co-cultured, and then implanted composite in vivo to repair sheep shin bone defects. Observing the effect of bone allograft with bone marrow stem cells to cure sheep defect, and the experiment as an experiment evidence in bone-tissue engineering.Extracting about5ml bone marrow from below the tibial tubercle of two1-month-old Mongolia Sheep, with sumianxin injection (0.1ml/kg) anesthesia. Bone Marrow-derived Mesenchymal Stem Cells was separated and purified from bone marrow. Researching the biological characteristics and osteogenic differentiation of bone marrow-derived mesenchymal Stem cells. Marrow-derived Mesenchymal Stem Cells adhered bone allograft co-cultured for24hours for use.9Mongolian Sheep, weighing50-60kg, under Sumianxin injection (0.lsml/kg) anesthesia and disinfection. We revealed two sides of shin bone middle and made1.0cm long bone-periosteum defect area. Random selected one side as experimental group, the other as control group, implanted the composite of bone marrow-derived mesenchymal stem cells combined and allogeneic bone in experimental group, and the other side repaired by allogeneic bone with nothing implants. After4,8,12week we randomly selected3sheep to observe the general situation, m-CT examination and histological examination.BMSCs isolated from Bone marrow can be cultured and purified with the method of density gradient centrifugation and adherence screening at the same time can proliferated rapidly in vitro. Under suitable condition, bone marrow mesenchymal stem cells can be induced to osteoblastand.Incubated the composite of Bone Marrow-derived Mesenchymal Stem Cells and allograft bone for different length of time (2hours,8hours,24hours,28hours) in vitro. Investigating the adhension of BMSCs on the allograft bone by scanning electron microscopy. Result display, BMSCs can grew in the both surface and the inner of bone. There is good biocompatibility bewteen BMSCs and allograft bone.4,8,12week After surgery, we randomly selected3sheep and killed them. CT and histological examination showed the composite of BMSCs with allograft bone could repair shin bone defects and the effect is significantly better than that in allograft.Conclusions:1. BMSCs isolated from Bone marrow can be cultured and purified with The method of density gradient centrifugation and adherence screening in vitro. BMSCs is the ideal seed Cells for bone defects in bone tissue engineering.2. There is good biocompatibility bewteen BMSCs and allograft bone. BMSCs with allograft bone could repair shin bone defects and the experiment could as an experiment evidence in clinical medicine.
