Jing Yuan Kang Capsule Fn Inhibition Of Extracellular Matrix Of The Bone Marrow Of Mice And Ln

Objective: Jing Yuan Kang Capsule observed inhibition of mouse femur extracellular matrix protein fibronectin (FN) and laminin (LN) in the bone marrow, explore Jing Yuan Kang Capsule improve chemotherapy-induced bone marrow suppression mechanism.Methods: Select56KM mice (male,6~8weeks age,18~22g weight),which were randomly divided into7groups (normal group, model group, Likejun tablet group, zhenqifuzheng Capsule group, high-dose Jingyuankang Capsule group, medium-dose Jingyuankang Capsule group, low-dose Jingyuankang Capsule group),8mice in each group. From the first day to the3rd day, every morning, the CTX(100mg/kg)was injected into the abdominal cavity of each mice, except themice of normal group, which were injected with0.9%NS. From the first dayto the seventh day, every afternoon, the mice of normal group and model group were infused with0.5%CMC-Na (20ml/kg), the mice of Likejun Tablet group were treated with Likejun Tablet (32mg/20ml/kg), the other groups were treated with zhenqifuzheng Capsule(3mg/20ml/kg) and Jingyuankang Capsule（4g/20ml/kg,3g/20ml/kg,2g/20ml/kg.)respectively. On the8th day, the mice were euthanized by cervical dislocation, then take out the bone marrow cell and disperse into separated cellular suspension on the ultra-clean work table. Take part ofcellular suspension to be used for the test of CyclinD1mRNA、CyclinEmRNA、CDK2mRNA and CDK4mRNA according to the kit instructions.Results:1by immunohistochemistry results showed that compared with the model group,each drug group could significantly improved laminin and fibronectin expression, which Jing Yuan Kang Lee High capsule group than the control group maybe the king piece, Zhenqi righting the control group.2PCR method Experimental results show that compared with the model group,the drug group could raise FNmRNA and LNmRNA gene expression levels, which Jing Yuan Kang high dose group was better than the other groups.Conclusion: Jing Yuan Kang Capsule can significantly improve bone marrow suppression mouse hematopoietic microenvironment cell morphology, increased expression of extracellular matrix FN and LN bone marrow cells, bone marrow s uppression LNmRNA and FNmRNA gene expression in mice, enhance foreignmarrow extracellular matrix proteins (laminin and fiber connexin) activity, promote the repair of hematopoietic microenvironment, thereby restoring and rebuilding hematopoietic function.
