Objective:This study aimed to examine the protective effects of Ferulic acid(FA) on A β1-42induced tau protein hyperphosphorylation on PC12cells and its possible mechanisms。Method:In this paper, PC12cells were treated with20μM Aβ1-42of absence or presence of FA in varying concentrations for24h. Then, cell viability was measured by MTT assay and LDH assay. Apoptosis of PC12cells was measured by Annexin-V/PI and casapse3activity assy. p-Tau(ser202、ser369)、T-tau、P-ATK (ser473)、AKT、P-GSK-3β(ser9)、GSK3β determined by western blotting.Result:The results showed that treatment with20μM AB1-42could induce cytotoxicity in PC12cells. However, different concentrations of FA(10.100and500μM) significantly increased the cell viability, decreased the MDA、ROS release, suppressed the apoptosis of PC12cells, up-regulated the P-AKT、 P-GSK3β protein expression compared with the AB1-42treated group, down-regulated the p-tau(ser202> ser369) protein expression compared with the Aβ1-42treated group.Conelusion:Ferulic acid protected PC12cells against Aβ-induced neurotoxicity by inhibition of Tau hyperphosphorylation through PI3K/AKT/GSK-3β signaling pathway and there maybe other protective mechanism related to oxidative stress and apoptosis.
