Purpose:Observation of Professor Ning Yagong in " Tonification and purgationin combination, adjust the balance"Method for the effect on islet function in type2diabetic rats recipe idea consisting of HuangDi JI-Xian decoction, To investigate therole of HuangDi JI-Xian on type2diabetes islet beta cells and its mechanism.Method: Choose SD male SPF60rats.10in the control group given normalsaline only, more than50only after the success of modeling,10rats in each groupwere randomly divided into:For the model group, physiological saline to westernmedicine group of metformin and different concentration of yellow Diji XianDecoction(high,lowdose group,medium).After8weeks of administration,measurementof body weight,Cardiac blood collection,The fasting blood glucose (FBG), fastingserum insulin (FINS), fasting C peptide(C-P) and calculate the Steady-state model ofislet β-cell function index (HBCI-β) and Steady-state model of insulin resistance(HOMA-IR), Simultaneous determination of rat triglyceride (TG), totalcholesterol(TC), C reactive protein. Finally, the rats were killed,access to aorta,Usinghematoxylin-Yi Hong (HE) staining was used to observe aorta endothelialmorphology,using immunohistochemical (SP) staining furtherobservation.Result:1.Weight:Body weight before and after treatment were observed and compared,themodel group was significantly decreased(P <0.01),metformin group, low, middledose group of Chinese medicine to reduce the level of no,Traditional Chinesemedicine high dose group was not significantly decreased.2.Serologically detected index:At the end of the experiment,compared with themodel group,In the traditional Chinese medicine, high dose group andmetformingroup rats fasting blood glucose (FPG), triglyceride (TG), total cholesterol (TC), C reactive protein (C-RP), Steady-state model of insulin resistance(HOMA-IR)decreased significantly (P <0.01or P <0.05),Fasting serum insulin (FINS), C peptide(C-P), the Steady-state model of islet β-cell function index (HBCI-β) increasedsignificantly (P <0.01or P <0.05).3.HE staining:Compared with the changes in vascular endothelial morphology inrats of model group,traditional Chinese medicine high dose group and the metformingroup were significantlyimproved; In the traditional Chinese medicine, small dosegroup had no obvious improvement.4.Immunohistochemical staining:Compared with the model group,TraditionalChinese medicine high dose group and the metformin group rat vascular endothelialmorphology was significantly improved,while the other group had no obviousimprovement of traditional Chinese medicine.Conclusion: Traditional Chinese medicine HuangDi JI-Xian decoction can reduceblood sugar, blood lipid levels in type2diabetic rats, significantly improve glucoseand lipid metabolism in diabetic rats, inflammatory reaction, increasing the seruminsulin levels and sulin sensitivity, thus improving islet function in type2diabeticrats.At the same time have protective effect on type2diabetic rat aortic vascularendothelial, in order to prevent the occurrence of Diabetes big blood vessel disease.
