| Objective:The development of CAD is related to genetic background and environment factors. The aim of this study is to explorethe effect of interaction between poly-miRTS and environment factors to the incidence of CAD, andprovide the theoretical basis for early screening of high risk population, primary prevention of CAD, and the early clinical intervention.Methods:The Koren-Chinese and Chinese healthy control (500) and patients with CAD(500) were selected from yanbianarea(1) PCR-RFLP strategy was conducted to study the ABCA gene polymorphism. this study.(2) The DNA was amplified by polymerase chain reaction(PCR), and the PCR product was cutted by restriction enzyme, the DNA was ligaded to pGL vector. The Ligation was trasnformed into E. coli for amplification of vector. The purified vector was transfected to human smooth muscle cell and the luciferase assay was performed.Results:CAD predisposing Poly-miRTS NAMPT rs1065322(p=0.003) ABCA1-rs4149341(p=0.0007) and CDKN2B-rs1063192(p=0.018) had significant differences between healthy control and patients. Only rs4149341(p=0.015) had significant difference between healthy control and patients in luciferase assay. Conclusion:ABCA1-rs4149341allele could be the predicting genes for CAD. These results provide the theoretical basis to develop the Single nucleotide polymorphism chip. Korean-Chinese CAD patients showed significant higher ABCA1-rs4149341G allele frequency than Han-Chinese. |
