Objective:To investigate the transcription factor KLF4on the proliferation of breast cancer cells to parthenolide and the related mechanisms.Methods:KLF4mRNA levels were detected by Real-time PCR in breast cancer cell lines. The expression of KLF4mRNA and protein in BT549cells was analyzed by Real-time PCR and Western blot analysis after pEGFP-KLF4transfection using LipofectaminTM2000; The expression of KLF4mRNA and protein in MCF-7cells was analyzed by Real-time PCR and Western blot analysis after siRNA-KLF4transfection using LipofectaminTM2000. Cell proliferation was measured by CCK8assay. The sensitivity of breast cancer cells to parthenolide was oberserved after KLF4overexpression or knockdown in breast cancer cells. P21expression was detected in MCF-7cells by cell immunofluorescence. Cell cycle was measured via flow measurement method.Results:(1) Compared with pEGFP-control group, BT549cells transfected with pEGFP-KLF4was significantly sensitive to parthenolide (P<0.05).(2) Compared with siRNA-negative control group, MCF-7cells transfected with siRNA-KLF4was lower sensitive to parthenolide (P<0.05).(3) Compared with the siRNA-negative control group, P21expression was significantly lower in MCF-7cells after parthenolide treatment for48h in siRNA-KLF4group by cell immunofluorescence.(4) Compared with pEGFP-control group, G0-G1cell cycle arrest was obervered in BT549cells transfected with pEGFP-KLF4after parthenolide treatment for48h (P <0.05); Compared with siRNA-negative control group, the percentage of G0-G1cell cycle decreased significantly in MCF-7cells transfected with siRNA-KLF4after parthenolide treatment for48h (P<0.05).Conclusion:KLF4expression levels in breast cancer cells was positively correlated with the sensitivety of parthenolide. BT549cells was higher sensitivity to parthenolide after KLF4overexpression; MCF-7cells was lower sensitivity to parthenolide after KLF4knockdown. It might be related with KLF4activating P21 protein and blocking cell cycle G1/S.
