Diabetes mellitus has become a hazard to the health of all peoples as a chronic disease. It can cause a variety of vascular disease complications such as cataract, nephropathy, neuropathy. The molecular mechanisms of vascular disease, include oxidative stress, the polyol pathway, protein kinase C activation, advanced glycation end products and hexosamine metabolic pathways and the like. Each pathway has not been fully elucidated clearly. Numerous studies have shown that these mechanisms lesions in certain metabolic processes often point contact with each other, so that different mechanisms influence each other, so that lesions incentives and consequences of increasing complexity. There was a consensus indicates that atherosclerosis is a kind of inflammation-based diseases. In diabetic patients, the incidence of lower limb arteriosclerosis is very high. Patients with poor hyperglycemic control, lower limb arteriosclerosis lead to foot ulcers, leg ulcers and even amputation example increasing year by year, which greatly threatens the quality of life and survival of patients. Mechanisms of diabetic vasculopathy understanded clearly is the fundamental way to improve the quality of life and cure with related complications.Ten years ago, has rediscovered short sequences of microRNA, with a variety of regulatory functions in normal human tissues and organs, while the vast majority of disease also plays a significant role, such as development and differentiation, proliferation control, and other aspects of the cycle and apoptosis. Especially its human blood can stably exist characteristics as a candidate for clinical diagnosis star molecules. With the in-depth study of microRNA gradually it is discovered microRNA process also involved in the pathogenesis of diabetic complications. Accordingly, the purpose of this study is to detect differences in microRNA expression in different circumstances the physical state of the patient’s blood in order to identify and offset relative to the normal expression of the microRNA, the establishment of diabetic lower limb arteriosclerosis differences in microRNA expression profiles lay the foundation for subsequent further study.This study successfully detected and identified three types of human blood samples and differential expression of microRNA data are diabetic lower extremity atherosclerosis in patients with non-diabetic patients with lower limb arteriosclerosis as well as healthy people without diabetes. According to the data, compare the different upregulated and downregulated microRNA, reflecting the state of the body in kinds of microRNA expression differences. This can also establish a small database for later in-depth study of the role of microRNA.
