[Objective]This study is established in the model of SD rats subtotal hepatectomy, based on the preoper-ative intermittent hypoxia in rats. After surgery, we detected the EPO and HGF levels of the re-sidual liver and the related indicators of the liver regeneration and features, by which the varia-tion of the EPO and HGF of the residual liver after subtotal hepatectomy could be explored. This study may provide more theoretical basis and technical path for relieving residual liver tissue damage and promoting liver regeneration by taking preoperative interventions.[Method]2.1. A toltle of120SD rats were randomly divided into three groups:Sham operation group(S group)、majority hepatectomy group(MH group) and Intermittent hypoxia precondi-tioning group (IHPC group). Blocking the hepatic blood flow for15mins, then the left and mid-dle liver lobes of MH group were resected The rats of IHPC group were exposed to hypoxic en-vironment of10%oxygen for lh/day. After7consecutive days, the left and middle lobes of liver were resected by the last hypoxixa(with MH group).2.2. Five rats of every group killed at2h,6h,12h,24h,48h,72h,120h and168h were tested. The lower vena cava blood was taken to do the liver function testing. The residual liver was weighted to calculate the degree of liver regeneration and regeneration index. Some of the resi-dual liver were used to detect the levels of EPO and HGF. Taking fresh liver tissue fixed by for-maldehyde and glutaraldehyde solid, we observed the ultrastructural changes under the transmis-sion electron microscope and the antigen expression of PCNA by immunohistochemistry.[Results]:The serum liver function values of IHPC group was located between the group and the S group, and a significant difference was between the groups. The liver tissue of three groups were observed by electron microscope. The hepatocytes of MH group were swollen, the mitochondria was severely swollen, which changed as a balloon, the matrix of which was enhanced and re-duced as floe. The mitochondria of IHPC group was mild swollen, the crest of which was more than MH group. The cell morphology was normal in S group.The level of PCNA in MH group is low, which showed a slow increasing trend at2h,6h and12h and significantly increased to reach a peak at72h, and then gradually declined. Compared with the MH group, the level of PCNA in IPHC group was significantly increased, particularly at48h,which reached a peak at72h and then gradually declined.The EPO level of postoperative residual liver tissue in the three groups had statistically sig-nificant difference. The level of EPO in IPHC group was between S group’s and MH group’s,which was observed in the trend pictures changed with time. The level in IPHC group was significantly higher than the MH group’s. The level of HGF in IPHC group was statistically higher than the MH group’s, but their variation was similar, they all reached the peak at24h and then gradually declined.[Conclusion]Intermittent hypoxia preconditioning can improve the levels of EPO and HGF in the rat re-sidual liver after hepatectomy, which promotes the residual liver regeneration and shows protec-tive effects on residual liver after partial hepatectomy.
