OBJECTIVETo detect the level of expression of transforming growth factor-β1(TGF-β1),Caspase-3and Wilms’ tumor suppressor gene (WT1) in the first renal biopsy tissuesof glomerulonephritis (GN). The result were discussed and analyzed on the meaningof pathologic change, clinical diagnosis and treatment in GN.METHODSThis study selected40cases of first renal biopsy tissues in patients with GN and7cases of control renal tissues and collected the clinical information of patients: Age,Sex, Course of disease, Serum creatinine (SCr), Total24hours urinary protein (TUpr),Plasma albumin (Alb), Pathological diagnosis, then divided the40cases of GNsamples into subgroups by pathological pattern, glomerular filtration rate (GFR) andTUpr. The expression of TGF-β1, Caspase-3and WT1in all samples of renal tissueswere examined by immunohistochemistry. Finally, it was analyzed that the meaningof expression of three marks among these subgroups and control group, relationshipof interacting among three ones, correlation between expression and clinical data.RESULT1.The expression of TGF-β1, Caspase-3, WT1were significant differences between40cases of GN and control group (t=6.256,P=0.000;t=4.744,P=0.000;t=-3.05,P=0.022). The expression of TGF-β1: The positive rates of TGF-β1in group ofMsPGN were lower than MN (t=-4.131,P=0.002); No significant differences in otherdivided groups. The expression of Caspase-3showed no differences in trial groups.The expression of WT1: Group of massive proteinuria were lower than thatnon-massive proteinuria (t=-2.130,P=0.046); No significant differences in otherdivided groups. 2. Expression of TGF-β1was showed negative correlation with TUpr and expressionof WT1(r=-0.373,P=0.033;r=-0.317,P=0.069), positive correlation with patho-logical pattern and expression Caspase-3(r=0.245,P=0.137;r=0.163,P=0.319) bymultiple linear regression equation. It was also showed that expression of WT1wasnegative correlated with Age, TUpr, expression of TGF-β1(r=-0.387,P=0.022;r=-0.325,P=0.098;r=-0.290,P=0.099). While expression of Caspase-3cannotestablish the equation.3. Cluster analysis assess correlation of clinical and pathological indexs, and it showsthat the complementary result with multiple linear regression equation;and it has highcorrelation among expression of TGF-β1, Caspase-3and WT1possibly, butexpression of Caspase-3has low correlation with span of patient courses.CONCLUSION1. TGF-β1, Caspase-3, WT1might play a synergistic action in early stage glomerulardiseases which will be developed to end-stage renal disease (ESRD). Up-regulatedexpression of TGF-β1and Caspase-3and down-regulated expression of WT1wereclosely related to pathologic impairment of glomerular.2. The positive rates of TGF-β1in group of MN were higher than MsPGN suggest thatexpression of TGF-β1may be relate to pathological pattern.3. Caspase-3have no significant differences in each trial groups, but it has highcorrelation with expression of TGF-β1, but low correlation with index of course.Possibly apoptosis is inhibited in mild or early GN diseases.4. Expression of WT1was negative correlated with Age, TUpr, expression of TGF-β1;while down-regulated expression of WT1was related to the decreased podocyte.
