ObjectiveTo investigate the Protective effect of Tongfu Xiere method of traditional Chinese medicine in mtDNA of liver cells about obstructive jaundice, and thus to protect and improve liver function about obstructive jaundice.MethodsThe studies includ animal studies and clinical studies.The clinical studies:The cases comply with the conditions in accordance with obstructive jaundice and non-obstructive jaundice were divided into experimental and control groups. Each case during surgery was excised liver tissue of about100mg, and stored at-80℃refrigerator for detecting test indicators, including HE stained tissue biopsy, MDA and8-OHdG. The control group were only treated with conventional therapy, in addition to the experimental group were treated with conventional therapy, as well as traditional Chinese medicine.The animal studies:In accordance with the principle of completely random, the10rabbits were divided into the control group1and the obstructive jaundice group, which is divided into the control group2and the TCM group. To establish an animal model of obstructive jaundice for the obstructive jaundice group rabbits, after removing the obstruction for10days, the rabbits of the TCM group were treated by Chinese medicine. According to different points of time of surgery, they were taken liver tissue in each group for each50mg, stored at-80℃refrigerator for experiments to detect targets, indicators, including MDA,8-OHdG. ResultsClinical research:The age, gender, renal function of the initial data in two groups of patients was no significant difference. But the ways of ALT, AST, GGT, ALB, TBIL, DBIL are statistically significant. HE staining of liver tissue results showed:the control group---the structure clear, normal liver cells arranged in a uniform radial, outline clear; obstructive jaundice group---disordered liver cable structure, bile pigment deposition in the liver cell cytoplasm. The quantitative detection of8-OHdG of mitochondrial about liver tissue suggested obstructive jaundice grouPwas significantly higher than the control group; MDA content in the obstructive jaundice group was much more than that in the control group.Animal studies Parts:The8-OHdG levels of the control group1was the least, and the TCM group was the highest. There were significant statistical difference between the TCM group and the control group2(P<0.01). MDA content of the control group2is much more than that of the TCM group, with statistically significant (P<0.05).ConelusionOur study clearly demonstrated that mtDNA damage is involved in liver damage in obstructive jaundice. The Cholagogic Xiaohuang Decoction participate in the treatment of obstructive jaundice, and one of the mechanisms is by which works to protect mtDNA. One of therapeutic mechanisms by Tongfu Xiere method is scavenging oxygen free radicals, protecting mtDNA and repairing damage mtDNA to protect liver cells and repair damaged liver cells.
