Objective:Chronic bacterial prostatitis is a common infectious disease in adultmales.Clinical manifestations of chronic bacterial prostatitis are diverse.Most of the patientsshow the pelvic pain, voiding dysfunction, sexual dysfunction and other symptoms.Available studies have shown that the recurrence rate is as high as20%.The current clinicaltherapy for chronic bacterial prostatitisgives priority to oral antibiotics, but the long-termcurative effect is not satisfying.Maybe the blood prostate barrier leads to the outcome.The existence of blood prostatebarrier can protect the prostate from the invasion ofharmful factors,howerver, at the same time, it makes the therapy drugs hard to enter theprostate local lesion, then difficult to reach the effective concentration.Therefore, how toincrease the drug permeability may be the key to the prostatitis treatment.Part1:The experimental use of escherichia coli to establish chronic bacterialprostatitis model in the rat.Objective:Explore suitable concentration of e. coli to establish the rat model of chronicprostatitis features,for the later research of the establishment of animal model.Method:Choose36healthy adult SD rats and randomly divide them into control group, lowconcentration, and high concentration group.Use the injection method to make an animalmodel with E. Coli. Inject the control group with physiological saline.The injectionconcentration in low concentration group is1.5x108cfu/ml fluid of e. coli, and theinjection concentration in high concentration group is3x109cfu/ml fluid of e. coli.After4weeks, kill the rats, and observe the pathological changes in the rat prostate.Use thehomogenate mechanism of prostate tissue,to culture the prostate gland homogenate colonyafter gram staining microscopy. Results:The control group rats prostate have no inflammatory manifestations. Theprostatic inflammation in the high concentration group is similar to acute suppurativeinfection, and has a high mortality rate. The rat prostate in the low concentration group issimilar to chronic inflammation, and conforms to the chronic prostatitis.Conclusion:The method injecting the e. coli liquid with the concentration of1.5x108cfu/ml can successfully establish a stable rat model of chronic bacterial prostatitis. Part2:The effect of low dose ultrasound mediated microbubble on the openingexperimentrat of blood prostate barrierObjective:Use Evans Blue as a tracerto research the effect of Low dose ultrasoundmediated microbubble on theopening of.blood prostate barrierMethod:Distribute48male healthy SD rats randomly into4groups,the control group,the EB group, immediate group, and delayed group.All rats are given the same dose ofultrasound irradiation for5min.Inject the control group with saline solution,according tothe weight50mg/kg dose, inject EB group with EB. According to the weight0.1ml/kgdose,inject microbubble group with microbubble,Give the delay group ultrasonicirradiation for5min after the injection with microbubble, and inject EB after30minutes.After the experimental, observe the prostate tissue and detect the concentration ofEB within the prostate.Results:Drug concentration of prostate tissue in EB groupis higher than that in controlgroup(P<0.05),while drug concentration of prostate tissue in immediate group is higherthan that in EB group and delayed group(P<0.05).Conclusion:Low dose ultrasound mediated microbubble contrastmedia can open bloodprostate barrier, improve drug concentration of prostate tissue, and have a certaintimeliness. Part3:The curative effect of Low dose ultrasound mediated microbubble onchronic bacterial prostatitis in a rat modelObjective:Use Low dose ultrasound mediated microbubble in the rat model to open the blood prostate barrier, increase the drug concentration within the prostate, and finallyget the treatment purpose.Method: Choose48SD rats models with chronic bacterial prostatitis and randomlydivide them into four groups, control group, drug group, microbubble group, drug+microbubble group.All rats are given the same dose of ultrasound irradiation for5min.According to the weight, inject the control group with0.1ml/kg dose of Physiologicalsaline.According to the weight,inject the drugs group with0.1mg/kg dose oflevofloxacin.According to the weight,inject the microbubble group with0.1ml/kg dose ofmicrobubble,andthe drugs+microbubble group with0.1ml/kg dose of levofloxacinmicrobubble mixture.Treat every two days and,last for eight days.After the experiment,observethe changes in the rat prostate pathological tissue and gradethepathology..Results:Compared with drug and microbubble groups, invasion glands with in theprostate gland inflammatory cells in drug+microbubble group decreases significantly, andhave statistical significance,P<0.05.Conclusion:Low dose ultrasound mediated microbubble can increase the drugconcentration of prostate and eradicate the prostate infection.
