The Association Between IL10, TNFSF15and Behcet’s Disease In Northern Chinese Han

Objective. Interleukin10(IL10) is a pleiotropic cytokine with complex and multiple effects in immune response. It has anti-inflammatory effects which are mediated by down-regulation of the Th1cytokines. Previous GWAS and candidate gene association studies had identified IL10as a risk locus for Behcet’s disease (BD). BD and Crohn’s disease(CD) shared similarities in their mechanism, clinical manifestations and treatments, especially intestinal BD and CD. The study aimed to investigate the association between IL10and Tumor necrosis factor receptor superfamily member15(TNFSF15) polymorphisms and BD in northern Chinese Han.Methods. Four hundred and seven BD patients and679healthy controls were enrolled, and genotyped by Sequenom MassArray system (Sequenom iPLEX assay, San Diego, CA).Results. The frequency of risk allele of rs1800871-T was notably higher in BD than that of controls (71.9%vs.66.2%, OR:1.30,95%CI:1.08-1.58, pc=0.036). Similarly, the frequency of risk allele of rs1518111-A was clearly higher in BD than that of controls (71.7%vs.66.1%, OR:1.30,95%CI:1.08-1.57, pc=0.039). Similarly, the haplotype (GACC) formed by rs1518111, rs3021094, rs3790622and rs1800871was related to BD (x2=7.61, pc=0.035). However, TNFSF15(rs6478108, rs4263839) were not in a linkage with BD in northern Chinese Han (pc>0.05). Results of the meta-analysis combined our data and previous studies evidenced that rs1800871and rs1518111were associated with BD (OR:1.41,95%CI:1.30-1.52, phet=0.193; OR:1.37,95%CI:1.28-1.47, phet=0.124).Conclusion. In a word, IL10could be regarded as the risk factor for BD in northern Chinese Han population. While TNFSF15(rs6478108, rs4263839) may be not associated with BD in northern Chinese Han.
