Objective:Discussion of amiloride hydrochloride on Interstitial Cells of cajal sample of the bladder (interstitial cells of cajal, ICCs) Ca2+impact cells.Methods:Isolation and Culture of ICC cells of bladder and bladder fluorescent identification of ICCs c-kit of cell membrane receptor; vitro ICC cells crawl after the success of the bladder, will be experimental cell crawling into the experimental group and placebo-treated group. Experimental group and100μmol/L,50u mol/L amiloride PBS solution treatment and placebo-treated group to PBS incubation. Under the microscope based on a typical cell morphology of urinary bladder of interstitial cells of Cajal-like (ICCs), random selection of20cells around the contactless ICCs of the bladder cells, isolated existence, using Fluo-4-AM after incubation in laser scanning confocal microscope.Results:50μmol/L amiloride group, ICCs in the cells of bladder Ca2+wave frequency slows down the statistics from before incubation tests no statistically significant (P>0.05), but Ca2+wave amPlitude significantly smaller than before, there was statistically significant (P<0.01),100μmol/L amiloride in the treatment group ICC cells of bladder Ca2+fluctuation of frequency and amplitude reduced significantly (P<0.01) and no statistically significant changes before and after the control group.Conclusion:Amiloride can reduce the ICCs in the cells of bladder Ca2+fluctuation of frequency and amplitude, it may be a potential drug treatment of OAB.
