[Objective] The experiment by removing the bilateral ovaries of SD rats, Establish SD rat animal model of low estrogen model, discusses low estrogen have been influenced to express of estrogen receptor alpha, matrix metalloproteinase9, matrix metalloproteinase2on SD rat TMJ condylar cartilage and KJ proximal tibial cartilage. In order to find out the differences and similarities between two kinds of articular cartilage.[Methods]16weeks old female SD rats were randomly divided into Experimental group and Control group, each group of16SD rats, Experimental group (surgical removal of the bilateral ovaries of SD rat), Control group (surgery to remove only the corresponding parts of the equal volume of fat). Respectively in SD rats were sacrificed after surgical2,4,6,8weeks, each group4SD rat, acquire TMJ condylar cartilage, KJ proximal tibial cartilage, observe Histological changes of TMJ condylar cartilage and KJ proximal tibial cartilage, immunohistochemical staining detection of different time points estrogen receptor a, matrix metalloproteinase2and matrix metalloproteinase9in SD rats in each group TMJ condylar cartilage and KJ proximal tibial cartilage expression changes by use of statistical analysis at each time point.[Results] Low estrogen model was effective. HE stain revealed TMJ condylar cartilage and KJ proximal tibial cartilage of the Experimental group happened pathological changes, and the TMJ condylar cartilage Experimental group After surgery of6weeks, was the most serious, the KJ proximal tibial cartilage Experimental group after surgery of8weeks, was the most serious. The expression of ERa in TMJ condylar cartilage Experimental group was lower than Control group, the expression of MMP-9and MMP-2was higher than Control group.The expression of ERa in KJ proximal tibial cartilage after stimulation of2weeks Experimental group was higher than Control group, but after stimulation of4,6,8weeks Experimental group was lower than Control group,the expression of MMP-9and MMP-2was higher than Control group[Conclusions] This rats model of the low estrogen shows great practicability and reproducibility. By removing the bilateral ovaries of SD rats has been effected on the expression of Era, MMP-9and MMP-2in TMJ condylar cartilage and KJ proximal tibial cartilage. This change may be caused by SD rats reasons for the changes of the temporomandibular joint, knee joint structure, and can also be caused the temporomandibular joint and knee joint disease pathology foundation.
