Expression And Adhibition Research Of D-PAS、IgG、IgM、PDC-E₂and CD11c In Autoimmune Liver Disease

[Objective]Researching the pathological features of autoimmune liver disease, finding effective indicators of pathological diagnosis.[Objective]Screening liver biopsy cases of2006to2012from department of Pathology of Yunnan Kunming Medical University.Choosing30cases of serum AMA-M2positive、17cases of serum AMA-M2negative which the pathological diagnosis are primary biliary cirrhosis and20cases of autoimmune hepatitis.Using D-PAS specific staining to understand bile duct lesions in portal area.Using the immunohistochemical method to detect the expression of IgG, IgM, PDC-E2, CD11c antibodies in tissues.[Results]1. D-PAS special stain can sketch the contours of the contours of biliary epithelial basement membrane,it is in favor of understanding the damage for bile ducts in portal area for the PBC patients.2. Immunohistochemistry revealed:IgM、IgG positive particles mainly distributed in the plasma cells cytoplasm of liver portal area. PDC-E2positive particles expressed in the cytoplasm of biliary epithelial cells and liver cells, CD11c positive particles expressed in the dendritic cell cytoplasm of liver portal area. IgM、IgG levels in PBC AMA-M2positive group were significant difference(P<0.05), IgM、IgG levels in PBC AMA-M2negative group were significant difference(P<0.05),IgM、IgG levels in AIH group were significant difference (P<0.05),PBC patients mainly express IgM in the plasma cells in liver portal area, AIH patients mainly express IgG in the plasma cells in liver portal area.PBC AMA-M2positive group compared with the AMA-M2negative group, IgM、IgG levels were significant difference (P<0.05), IgM and IgG positive cells in PBC AMA-M2-positive patients are more than the PBC AMA-M2-negative patients.PBC AMA-M2positive group, PBC AMA-M2negative group, AIH group compared pairwise,PDC-E2antibody-positive units were significant difference(P <0.05);PBC AMA-M2positive group, PBC AMA-M2negative group, AIH group compared pairwise,CD11c positive units were significant difference(P<0.05) except comparison between PBC AMA-M2positive group and PBC AMA-M2negative group.[Conclusions]1.D-PAS special stain can understand the damage in bile duct basement membrane in liver portal area,and more intuitively perform the number changes of small bile ducts.2..PBC patients mainly express IgM in the plasma cells in liver portal area, AIH patients mainly express IgG in the plasma cells in liver portal area. Analysing IgM and IgG in liver tissue contribute to AILD diagnosis.IgM and IgG positive cells in PBC AMA-M2-positive patients are more than the PBC AMA-M2-negative patients.3.Liver biopsy tissue PDC-E2immunohistochemistry can help PBC diagnosis, PDC-E2expression have certain differences between PBC AMA-M2positive group and PBC AMA-M2negative group.4.CDllc mainly express in the liver portal area granuloma, it can help PBC diagnosis.
