[Objective] The relationship between NDRG1and colorectal cancer has different opinion in recent years. Aim to explore the differences of NDRG1gene between Yunnan human colon cancer cells with Caucasians of colon cancer cells. Expression difference of NDRG1was observed at the mRNA level.[Methods] Ascites of Yunnan colorectal cancer patient was collected, and isolated cells were cultured and passaged. NDRG1gene primers were designed. Total RNA was extracted from the colorectal cancer cells and cDNA was obtained by reverse transcriptase PCR, followed by agarose gel electrophoresis and DNA sequencing. Its molecular characteristics and functional properties were analysed bioinformatonally, according to Yunnan human NDRG1gene. Expression of NDRG1gene was observed at the mRNA levels in colorectal cancer cell lines of the Caucasians and the cultured ascites cells of Yunnan human colorectal cancer by real-time PCR.[Results] The cDNA band around1185bp, i.e. the NDRG1gene molecule size, was showed by Agarose gel electrophoresis. The reference nucleic acid sequence of NDRG1was that released by http://www.ncbi.nlm.nih.gov/nuccore/NM001135242.1Through bidirectional sequencing, we identified the sequence obtained from the Yunnan human colorectal cancer NDRG1is full sequence, with an open reading frame length1185bp, encoding394amino acids. Using NCBI BLAST software to do online comparison, it is showed that our sequeces were exactly the same as the online upload NDRG1nucleic acid sequences. The data showed that there was no difference of NDRG1in Yunnan human colon cancer cells and Caucasians colon cancer cells at the nucleic acid sequence level. Quantitative PCR data showed high levels of NDRG1in Yunnan colorectal cancer.[Conclusions] There is no difference of NDRG1gene sequences but mRNA expression level.
