Correlation Study Between NO、Immune Factors And Pathological Pregnancy

ObjectiveTo investigate relationship between levels of Nitric Oxide(NO),normaland abnormal pregnancy relativity between NO concentration and cytokines,therole of natural killer cell activity and T cell subset in the pathologicalpregnancy,thechanges of NO and celluar immunity function around tyeatmentwith Chinese medicine to the pathologic pregnant,the relation between placentalcell apoptosis and abnormal ppregnancy.MethodsConcentration of NO in placente and serum were done in normalpregnant(women),nopregnant,spontaneous abortion,dead fetus,fetal growthretardation by nitric acid reductase;levels of IL-6, IL-8, IL-10and TNF in serumof the patients with recurrent spontaneous abortion(RSA) were detected byELISA;activity of NK cell of the patients with dead fetus and RSA were studiedby MTT;with SAP method T cell subset changes were determined in the patientswith dead fetus and RSA;placental cell apoptosis were observed with nuclearfast red dyeing.Resultslevels of NO was higher in early,middle and late period of normalpregnancy,than nonpregnant women,The concentration of NO in meddle periodwas the most significant,tending to decrease in late period.Nitric Oxide levelswere much higher in RSA and dead fetus women,while NO levels in intrauterinegrowth retardation decreased remarkedly.Contents of IL-6, IL-8,TNF were significenty defference compared tocontrals(p<0.01).There was lower levels of IL-10.Activity of NK cells were raised,CD8cells lowed,proportion of CD4to CD8increased,the number ofcellular apoptosis in placenta increased in the patients with RSA treated byChinese medicine.ConclusionApropriate levels of NO and cytokines in the body play a definit role inkeeping normal pregnancy.Activity of NK cells and proportion of CD4to CD8participate in the pathological mechnisim of RSA and dead fetus;abnormalpregnancy cansed by NO or cytokines were possibly related to cellular apoptosisof placenta;more successful opportunity of pregnancy were prorided withChinese Medicine treatiment.
