ObjectiveTo investigate the expression of IL-27and its correlations with Th1, Th17inaplastic anemia.Material and MethodsPlasma levels of IL-27, IFN-γ, IL-17were determined by ELISA; Frequency ofTh1、Th17in PMBCs were detected by flow cytometry; and mRNA exression ofIL-27、IFN-γ、T-bet、IL-17、RORγt in PMBCs were quantified by real-time RT-PCR.In addition, the effect of exogenous rhIL-27on the differentiation of Th1、Th17wereassayed by in vitro cell culture experiments.Results1Plasma level of IL-27and mRNA expression of IL-27in PBMCs from patientswith AA (SAA131.65±36.16pg/ml、CAA105.76±23.75pg/ml) were significantlyhigher than that of normal controls (86.46±10.78pg/ml,P<0.05);2Plasma levels of IFN-γ and IL-17were significantly higher in AA (IFN-γ:SAA46.61±8.84pg/ml, CAA42.42±6.71pg/ml; IL-17: SAA36.85±8.60pg/ml,CAA32.59±11.16pg/ml) than that of normal controls (IFN-γ6.77±5.03pg/ml;IL-1718.64±4.08pg/ml). A positive correlation was found between plasma levelsof IL27and IFN-γ (SAA r=0.518, P=0.033; CAA r=0.588, P=0.006). However,no correlation was found between plasma levels of IL-27and IL-17(P>0.05).3Expression levels of Th1(SAA19.68±4.20%;CAA15.12±5.28%)、Th17(SAA2.03±0.85%; CAA1.86±0.43%) were significantly higher in patients withAA than that of normal controls (Th110.03±3.39%; Th171.34±0.40%). Plasmalevels of IL-27correlated positively with frequencies of Th1in AA patients (SAA r=0.599, P=0.011; CAA r=0.500, P=0.025). No correlation was found between plasma levels of IL-27and expression of Th17in patients with AA (P>0.05).4Exogenous rhIL-27could upregulate the expression of Th1significantly, inparallel with a remarkably upregulation of T-bet and IFN-γ mRNA expression.Expression of IL-17, Th17and its specific transcription factor RORγt were notaffected statistically in the presence of exogenous rhIL-27.ConclusionThe expression of IL-27, Th1and Th17were significantly elevated in AA. Theupregulated IL-27might contribute to the pathophysiology of immunologicderangement in patients with AA.
