Object:To investing the mechanism of Epithelial growth factor (EGF) in promoting diabetic foot ulcers (DFU) wound healing.Methods:To set up the animal model of diabetic foot,we choose32New Zealand rabbits from70New Zealand rabbits in the standard of blood sugar more than11.1mol/L.32New Zealand rabbits were used to make the models of DFU, then they were randomly assigned to two groups, EGF group (group A) and control group (group B),16for each group. The all layers skin of two groups animal were removed in rectangle area of10mm X7mm.After15days,we taked out new flesh organize though2mm margin of diabetic foot ulcers.Tissue sections of each DFU were masson and HEtrichrome stained after15days and observed under electron microscope. EGF mRNA were detected usingRT-PCR.Results:(1) Masson staining:Comparing group A with group B, group A healed better than group B, fibroblasts and vascular were more abundant in group A than group B.(2) Electron microscope:the fibroblasts of group A were bigger and in good shape, organelle were rich cytoplasm. A great amount of collagen were very regular surrounded the cells. For group B, the fibroblasts were irregular shaped, collagen were irregular surrounded the cells, few organelles were presented in the cells.(3) EGF mRNA detection:the expression of EGF mRNA was significantly increased in group A (p<0.05).Conclusions:EGF can promote the wound healing of DFU, possibly EGF is acting on epithelial cells and fibroblasts and thereby promoting the restoration of damaged epithelium. Moreover, external EGF might stimulate the expression of internal EGF.
