Objective:Study on the expression of Cathepsin C in cuprizone-induceddemyelinated model of central nervus system(CNS) in mice and discuss its clinicalsignificance.Methods:In animal model,six weeks old male C57BL/6mice were treated0.2%cuprizone for2weeks and5weeks in order to creat demyelinating model of CNS,Normal diet C57BL/6mice were used for blank control group.Myelin basicprotein(MBP) immunohistochemistry(IHC) staining and Black Gold Staining were usedto observe the normal myelin and remaining myelin of mice which treated0.2%cuprizone for2weeks and5weeks;Iba-1IHC staining was used to analysis theaccumulation of microglia/macrophage lineage cells;Catheosin C respectively withMPO/Iba-1/GFAP immunoflurescent double staining was used for expression andcellular localization of Cathepsin C.In clinical experiment,we use enzyme-linkedimmunosorbent assay(ELISA) to observe the level of Cathepsin C in cerebrospinalfluid(CSF) of multiple sclerosis(MS).Results:1.According to the results of MBP IHC Staining and Black Gold Staining,Wefound demyelination occurred in corpus callosum following exposure to cuprizonecompared with control group,and demyelination in5weeks group was more seriousthan that in2weeks group. 2.In control group,Cathepsin C was not expressed and there are a littlemicroglia in corpus callosum.After cuprizone treated,the number of microglia/macrophage lineage cells and the expression of Cathepsin C in5weeks groupwere higher than2weeks group.3.Iba-1/MPO/GFAP Immunofluorescent double staining showed that Cathepsin Cwhich detected in corpus callosum was expressed mostly by microglia and partly byinfiltrating neutrophils in CNS.4.The levels in MS patients’CSF were obvious higher than other neural diseasespatients.Conclusions:1.Demyelination occurred in corpus callosum following exposure to cuprizone.2.The accumulation of microglia/macrophage lineage cells which was induced byupregulated Cathepsin C and infiltration of neutrophil would be involved in aggravateddemyelination in C57BL/6mice.3.It indicated that Cathepsin C might participate in the pathogenesis of MS.
