| Background:Human platelet antigens (HPA) are immunogenic transmembrane glycoprotein antigens expressed on the surface of platelets.Human platelet antigens incompatibility can cause homoimmune reaction and produce platelet alloantibody, causing alloimmune thrombocytopenia, evoking a series of clinical syndromes such as platelet transfusion refractoriness (PTR), post transfusion thrombocytopenic purpura (PTP),neonatal alloimmune thrombocytopenic purpura (NAITP),etc. HPA incompatibility even can cause graft rejection in clinical transplantation. So identification of human platelet antigens have significance for clinical and blood transfusion.Objective:To investigate the genetic polymorphism of HPA-1to-17antigens in Anhui Han population and to predict the possibility of homoimmune reaction by HPA in Anhui region,so as to provide experimental basis to screen compatible platelets for clinical and to establish an HPA-typed platelet donor registry preliminary.Methods:A total of150samples from Anhui Han blood donors were randomly selected and collected2ml whole blood respectively, DNA was extracted from whole blood and genotyped HPA1-17by sequence specific primer polymerase chain reaction(SSP-PCR), Then the HPA genotype frequencies, gene frequencies and mismatch probability in random platelet transfusion were calculated.Results:The gene frequencies of HPA-1a,2a,3a,4a,5a,6a,15a were0.9966,0.8800,0.6267,0.9966,0.9700,0.9533and0.4833respectively in Anhui Han population, HPA-1b,2b,3b,4b,5b,6b,15b were0.0034,0.1200,0.3733,0.0034,0.0300,0.0467,0.5167. The gene frequencies of HPA-7a,8a,9a,10a,11a,12a,13a,14a,16a,17a were1.0000,HPA-7b,8b,9b,10b,11b,12b,13b,14b,16b,17b alleles were not detected. Mismatch probability in random platelet transfusion by HPA-1-. HPA-2, HPA-3, HPA-4, HPA-5, HPA-6. HPA-15were0.00675,0.1889,0.35843,0.00675,0.0565,0.08507,0.37472respectively.Conclusions:HPA1-17in Anhui Han population shows its own characteristics, HPA-2, HPA-3, HPA-15are highly polymorphic, Mismatch probability of random platelet transfusion from high to low are caused by HPA-15, HPA-3, HPA-2. |
