Objective To explore the expression changes of NOX2and NOX4in the cerebraltissue of patients who suffered from traumatic brain injury (TBI), and to analyze thecorrelation between the expression variety of NOX2and NOX4and traumatic brain injuryseverity, injured time and prognosis of the patients.Method30specimens of brain tissue within1cm around the contusion werecollected from the patients who suffered from TBI during the operation. The specimens ofbrain tissue were embedded with paraffin after dehydration and fixation and clipped. Theexpression level of NOX2and NOX4in neurons and astrocytes was detected withimmunofluorescence double staining. The groups were divided according to the injuredtime, glasgow coma scale (GCS) and glasgow outcome scale (GOS). The correlationbetween the expression variety and prognosis of the patients was researched. All data wereanalyzed with statistics.Results(1) The expressions of NOX2in neurons reached peak at12h-24h and the expressionsof NOX4in neurons reached peak at24h-48h after trauma. The time of theexpressions of NOX4arriving at peak was later than that of NOX2.(2) The expressions of NOX2in astrocytes reached peak at6h-12h and then reducedwith time. The expressions of NOX4in astrocytes were lower in the early time andreached lowest at6h-12h after trauma. Then the expressions of NOX4increasedgradually with time.(3) The GCS of patients was higher and the expression level of NOX2and NOX4inneurons was lower. (4) The GCS of patients was higher and the expression level of NOX4in astrocyteswas lower. The expression level of NOX2in astrocytes had no statisticalsignificance.(5) The GOS of patients was higher and the expression level of NOX2in astrocyteswas lower.(6) The expression level of NOX2and NOX4had no statistical significance in thepatients who suffered from cerebral hernia or not.Conclusions(1) The changes in the expression level of NOX2and NOX4in cerebral tissue within48h after trauma suggest the mechanism of oxidative stress (OS) play an importantrole in TBI.(2) In neurons, the parallel rise of the expression of NOX2and NOX4suggests themplay an important role in the early time after trauma. But the peak time of theexpression of NOX4is later and the suggestion is NOX4play an important role inthe later period after trauma.(3) In astrocytes, the expression of NOX2and NOX4has inverse relationship indifferent period after trauma. These imply that NOX4may have a protective effect.(4) The expression level of NOX2and NOX4in cerebral tissue is correlated with GCSand GOS of patients. These imply that the expression of NOX2and NOX4incerebral tissue can be used as a reference index to judge the degree of brain injuryand prognosis...
