Objective:To investigate the protective effect of black barley pericarp extract postconditioning on cardiomyocytes against hypoxia/reoxygenation (H/R) injury and to elucidate its underlying mechanism.Methods:Hypoxia/reoxygenation injury in H9c2cells was induced by Na2S2O4. H9c2cells were randomly divided into5groups:normal controls (no treatment), H/R, and H/R followed by drug treatment with low/median/high dose(H/R+L/M/H). The relative activity of cells was measured by MTT assay. Cell apoptosis, the generation of intracellular ROS and△Ψm were monitored by flow cytometer. The protein levels of Akt, pAkt, Bcl-2and Bax in H9c2cells were observed by Western blot assay. Statistical Analysis SAS9.2and SPSS19.0were used for statistical analysis.Results:1. Hypoxia/reoxygenation model (exposed for20min to5mM Na2S2O4for hypoxia, followed by reoxygenation for8h) proved to be stable and reliable.2. Hypoxia/reoxygenation decreased the relative activity of H9c2cells. Black barley pericarp extract postconditioning could inhibit significantly the degression of the relative activity of H9c2cells.3. Hypoxia/reoxygenation induced the apoptosis of H9c2cells. Black barley pericarp extract postconditioning could inhibit significantly the apoptosis of H9c2cells.4. Hypoxia/reoxygenation increase the lever of ROS in H9c2cells. Black barley pericarp extract postconditioning could inhibit significantly the augmentation of the lever of ROS in H9c2 cells.5. Hypoxia/reoxygenation decreased the level of△Ψm in H9c2cells. Black barley pericarp extract postconditioning could inhibit significantly the degression of level of△Ψm in H9c2cells.6. Hypoxia/reoxygenation decreased the expression of pAkt and Bcl-2in H9c2cells, Black barley pericarp extract postconditioning could inhibit significantly the depression of expression of pAkt and Bcl-2in H9c2cells, there was no evident change among the five groups in expression of Akt and Bax.Conclusion:Black barley pericarp extract postconditioning could protect H9c2cells against hypoxia/reoxygenation injury may via inhibiting the production of ROS, stabilizing△Ψm and inhibiting the apoptosis, which may relate to activation of the PI3K/Akt/Bcl-2pathway.
