The Expression Of Protein Arginine Methytransferase6and Its Significance In Lung Tissues Of Chronic Obstructive Pulmonary Disease Patient

Objective:The paper aimed to observe the levels of PRMT6mRNA, IL-13mRNA, COX II mRNA and the changes in histone H3K4me3and H3R2me2a signal in patients of lung tissues with COPD, and discuss the expression and significance of PRMT6in tissue of COPD patients.Methods:According to the latest COPD diagnostic criteria,31patients underwent pneumonectomy has been divided into3groups:10no-smoking and without CODP,10smokers without COPD, and11COPD group (stable stage). QRT-PCR has been used to measure PRMT6mRNA and inflammatory genes expression. Western-blot has been used to measure PRMT6protein and signal in histone H3K4me3and H3R2me2a.Results:1.The differences of FEV1%pre, FEV1/FVC%and PEF%pre between smokers without COPD and no-smoking without CODP are not significant (p>0.05). FEV1%pre, FEV1/FVC%and PEF%pre in COPD patients are significantly lower than those in smokers without COPD and normal controls (p<0.01).2. Compared with the no-smoking without CODP group, PRMT6mRNA expression decrease (p<0.05), but inflammatory (IL-13, COX-2) genes mRNA expression increase significantly in smokers without COPD group and COPD group (p<0.05).3. Compared to the no-smoking without CODP group, PRMT6 protein expression decrease in healthy smokers group and COPD patients group (p<0.05), and histone H3K4me3and H3R2me2a signal decrease in smokers without COPD group and COPD group.Conclusions:Protein arginine methyltransferase PRMT6protein expression decreased. H3R2me2a signal level drop, and the histone H3K4methylation level increase,but the inflammatory cytokines IL-13, cox-2mRNA expression increase to participate in the formation of COPD.
