Objects:The aim of our study is to investigate that the influence of mesalazine or prednisone on expression of intestinal mucosa CGRP and TNF-a level of mice with Dextran sulphate sodium (DSS) induced Experimental colitis.Methods:40specific pathogen free, about8week-old female BALB/c mice (weight20.0±2.0g), were randomly divided into4groups: normal, model, mesalazine group, prednisone group.10mice were involved in each group. Colitis was induced by administration of4%DSS (MW:40000g/mol) in drinking water to mice for7days. From the next day, each experimental group was administrated vehicle (distilled water), mesalazine (0.4g/kg), prednisone(5mg/kg) by gavage, respectively. During the observation, active state, mental state, loss of weight, stool consistency and occurrence of gross blood in stool were recorded. According to the mean DAI score、the length of colon from the colocecal junction to the anus, the efficacy of colitis was validated. After mesalazine or prednisone treatment, the express of TNF-α、CGRP in colonic tissue is assayed by immunohistochemical staining.Results:1.In model group,the mount of CGRP decreased,TNF-a increased,different results show in treated groups, these differences have statistical significance.2.with CGRP increases,The level of TNF-a decreases. Conclusion:In each group, we can see the phenomenon that CGRP increased, TNF-a decreased.
