Tissue factor (TF) is an important factor of physiological hemostasis, which exert hemostatic function through trigger the extrinsic coagulation pathway directly and amplify the intrinsic coagulation pathway indirectly. In this study, we will observe the influence of TF on the expression of P-selectin(Ps) in endothelial cells, and the inhibition of tissue factor pathway inhibitor(TFPI).AIM:The purposes of this study were to observe the effects and mechanism of tissue factor(TF) on the expression of P-selectin in human umbilical vein endothelium derived cell line (HUVEC)s. And to explore the effects of tissue factor pathway inhibitor(TFPI) on the expression of P-selectin (Ps) induced by TF.Methods:HUVECs and THP-1were both cultured in DMEM. HUVECs activity was determined with MTT chromometry. The adherence rate was determined with chromometry. Ps antigen was measured by ELISA Kit. Ps mRNA was examined by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR).Results: ①Compared with the control, a gradual increase in Ps antigen(r=0.958, P<0.05), Ps mRNA(P<0.05) and the adherence rate (r=0.972, P<0.05)were observed in HUVECs stimulated with increasing concentration of TF(1、10、50、100、150pg/ml), and peaked at100pg/ml.②TFPI(1、10、50、100、150ng/ml) alone could not affect the adherence rate of HUVECs(P>0.05). When pretreated with TFPI(1.10.50.100.150ng/ml) in30min, it inhibited the effects of TF on Ps antigen (r=-0.946, P<0.05), mRNA (P<0.05) and the adherence rate(r=-0.934, P<0.05) in dose-dependent manner, and100ng/ml was the strongest concentration.③Compared with the control, PDTC, which is the inhibitor of NF-κB pathway, had marked effects on Ps antigen expression induced by TF at antigen level as well as mRNA(P<0.05).Conclusion:The present data suggested that①TF could induce the expression of Ps in vascular endothelial cells with dose-dependent manner and time-dependent manner.②TFPI could inhibit the expression of Ps induced by TF.③TF can increase HUVECs adherence rate, and TFPI depressing this affect.④NF-κB translocation participate in TF-induced Ps expression.
