RPE cells transfecteby adenovirus-mediated Slit2(Ad-slit2),then culturaled12h、24h、48h.3.Before transfected by Ad-slit2,RPE cells were pretreated with MEKinhibitor PD98059、PI3K inhibitor LY294002for1h, then cultured12h、24h、48h.4.VEGF mRNA expression levels were detected by quantitativePCR and the protein expression levels were detected by ELISA.The proteinexpression levels of t-ERK、t-AKT、p-ERK、p-AKT were detected byWestern blot.Results: VEGF mRNA and protein expression levels were obviously increasedin RPE cells transfected with Ad-slit2at48h(P<0.05),p-ERK、p-AKTprotein expression levels were increased in RPE cells transfected withAd-slit2(P<0.05),but t-ERK and t-AKT remained unchanged,PD98059decreased the activation of p-ERK and LY294002decreased the activationof p-AKT,VEGF mRNA and protein expression levels were decreased byPD98059and LY294002(P<0.05).Conclusion:The expression of the VEGF in RPE cells transfected with Ad-slit2was increased.by MEK/ERK and P13K/Akt signaling pathways. | Keywords/Search Tags: | Slit2gene, AKT, ERK, VEGF | PDF Full Text Request | Related items |
